It promotes natural killer (NK) cell expansion and enhances intrinsic NK cell cytotoxicity. Two polymorphisms, -330T/G and+114T/G, have been identified in the IL-2 gene and are associated with various diseases and cancers. The aim of the study is to study the expression and to assess the impact of genetic polymorphisms of IL-2 on the susceptibility and prognosis of non-Hodgkin's lymphoma (NHL). Sixty patients with NHL as well as 60 age and sex matched healthy control subjects are included in this study. IL-2-330 T/G and+114 T/G genotypes were determined by PCR-RFLP. Our study revealed that both IL-2-330 T/G and+114 T/G gene polymorphisms are associated with increased risk of developing NHL with OR=3.609 (95% CI=1.527-8.417) and 4.142 (95% CI=1.637-10.538) respectively. Also, they increase the risk of bad outcome with OR=17.300 (95% CI=3.392-87.725) and 10.424 (95% CI=1.870-58.413) respectively. Moreover, combined presence of both polymorphisms is associated with about 6 folds increased risk of developing NHL. In conclusion, IL-2-330 T/G and +114 T/G gene polymorphisms could be involved in the pathophysiology and development of NHL. They may be useful as predictive molecular markers for prognosis and disease outcome in NHL patients ER -
