MicroRNAs (miRNAs) are small, non-coding RNAs control the expression of a number of genes involved in commitment and differentiation of hematopoietic stem cells and tumorigenesis. The study aimed to clarify role of miR-155,mir-424,in pathogenesis ,prognosis of AML and correlation of both miRNAs with NPM1 gene .The present study done on 40 adult newly diagnosed AML cases that were taken from National Cancer Inistitute & medical oncology unit Kasr Aini School of Medicine, Cairo University and 40 sex & age matched controls. A quantitative reverse-transcription PCR (qRT-PCR) assay was done to relatively quantify miR-424,miR-155. (RT-PCR) assay was done to assess NPM1 mutation and correlate it with both micro-RNAs.There was highly statistically significant difference in miR-424 between both patients and controls (p=0.0001),miR 155 was a bad predictor for AML cases. No correlation was found between miR -424, miR-155 and NPM1 .So that deregulated expression of both miRNAs does not depend on NPM1 to exhibit its role in leukemogenesis UR - http://172.23.153.220/th.pdf ER -
