Introduction : Renal ischemia reperfusion (IRI) injury is one of the major causes of acute kidney injury (AKI). The relationship between kidney and thyroid function has been well known for many years. Poly(ADP-ribose) polymerase (PARP) is a nuclear protein that is important for DNA repair, but its excessive activation is important in necrotic cell death. It remains unknown whether there is a relationship between thyroid hormones and PARP-1 expression during renal IRI or not.Aim: To elucidate the role of the thyroid hormones preconditioning in renal ischemia reperfusion injury and its possible role in the regulation of the PARP-1 activity. We also considered the pathological conditions of thyroid hormones (hypo and hyperthyroidism) and their effects on renal ischemia reperfusion injury.Methods: A total of 50 male albino rats 180-200 gm were designated randomly into the following five groups (10 rats in each group);group Ї: sham operated for renal IRI (Sham), group ЇЇ: (Euthyroid+IRI), group ЇЇЇ: (Hyperthyroidism+IRI), group ЇV: (preconditioned+IRI) and group V: (Hypothyroidism+IRI).Prior to surgery the thyroid state was confirmed by analysis of thyroid profile (T3, T4 and TSH) for all animals.At the end of the experimental period, systolic& diastolic blood pressure was measured, 24h urine was collected and body weight was measured for GFR determination. Blood samples were collected for assessment of angiotensin II, BUN and plasma creatinine. Tissue samples from the kidney for histopathological examination and for measuring the following parameters: malondialdehyde (MDA), superoxide dismutase (SOD), neutrophil gelatinase-associated lipocalin (NGAL), activating protein 1 (AP-1), Caspase 3, ATP, poly(ADP-ribose) polymerase (PARP-1) activity in the kidney
