TNBC forms 15% of breast cancer cases. It has a poor prognosis and lacks the responsiveness to hormonal and targeted therapies. Anthracyclines remain the most appropriate treatment for TNBC, but its cardiotoxicity necessitates selection of patients who will really benefit from it. From predictive factors of adjuvant anthracyclines sensitivity: Chromosome 17 CEP polysomy, TOP2A gene abnormality (deletion or amplification) and high Ki-67 proliferation index. Aim of work: To investigate TOP2A gene copy number and chromosome 17 CEP polysomy by FISH technique. Material aAnd methods: Immunohistochemistry for detection of Ki-67 LI and FISH technique for detection of TOP2A gene copy number and chromosome 17 CEP status, both were done on our study cases (50 cases) diagnosed at the pathology department, National cancer institute, Cairo-university. Results: CEP 17 polysomy showed a significant association with TOP2A abnormalities, and a trend towards better prognosis in patients with these abnormalities, So making studies on larger sample size may give significant association. Disease outcome, status of the disease (either locally advanced or advanced) and radiotherapy intake showed a significant association with overall survival.
