Effects of estrogen and angiotensin receptor blocker on cognitive impairment in ovariectomized rats /

The present study aimed to investigate the possible effects of treatment with telmisartan and 17Ý-estradiol (E2) alone or in combination on Alzheimers disease (AD) like symptoms induced by D-galactose administration to ovariectomized rats. Treatment with telmisartane and E2 alone or in combination improved animals behavior assessed by morris water maze and novel object recognition tests, alleviated histopathological alterations and reduced the level of inflammatory biomarkers 2tumor necrosis factor-Ü (TNF-Ü) and interleukin-1Ý (IL-1Ý)3 as well as AD biomarkers 2amyloid beta 1-42 (AÝ1-42) and acetylcholine esterase (AchE)3. This was associated with modulation in renin angiotensin system (RAS) favoring alternative neuroprotective arm (ACE2/ Ang (1-7)/ Mas receptor axis). Moreover, combined therapy produced more pronounced reduction in inflammatory biomarkers as well as AD biomarkers and more modulation of RAS than usage of each drug alone. These findings suggest possible therapeutic options for protection and delay of AD neurotoxicity
