Objectives: This in vitro study evaluated the effect of Essential oils (EOs), Essential oils with sodium fluoride (EOF) and Chlorhexidine (CHX) mouth washes on viability and apoptosis of fibroblast cell line. Material and Methods: Baby hamster kidney -21 (BHK-21) fibroblast cell line was cultured and incubated in Eagle's Minimum Essential Medium (EMEM) containing, EOs, EOF and CHX with different doses and various exposure times. Control cells received an equal volume of EMEM without adding mouth rinses. Cell viability and apoptosis were evaluated using MTT assay and RT-PCR. Results: The EOs, EOF and CHX mouthwashes inhibited fibroblast cell viability in a dose and time dependant manner. Conclusions: CHX mouthwash was the most cytotoxic to fibroblasts as compared to EOs and EOF. Moreover, adding fluoride to EOs in low concentrations didn`t worsen the adverse effects of EOs on cell viability and apoptosis induction
