Pasteurella multocida was isolated from nasal swabs of diseased animal , apparently healthy animal and lung sample of diseased and emergency slautered animal by three method one was by conventional ordinary culture method and the other were By applied for Two non-culturable methods depending on direct molecular detection on either examined sample or 24 hours preselective broth. Percent of isolation greatly different in each method as in conventional method culture percent were 9.3% from apparently healthy., 15.3% from pneumonic lungs of slaughtered animals, and 12 % nasal swabs of diseased animals with total recovery rate 13.0 %. While the Direct-PCR approaches While, by the Direct-PCR approaches. The direct PCR on samples detection percent were 16.0 % ,13.3 % & 22.0% from nasal swabs of healthy , diseased and pneumonic lung tissues, respectively .And direct PCR on preselective broth detection percent were (12.0%) from apparently healthy, (24.0%) from diseased animals and( 48.6%) from emergency slaughtered sheep and it is the highest detection rate (33.3%) of P. multocida in comparison to other two methods
