Background : Breast cancer (BC) is a major cause of cancer-related death worldwide. miRNAs have been demonstrated to play an important role in carcinogenesis either by oncogenic or tumor suppressor function.The aim of the present work : is to study the serum levels of miR-21& miR-10b to evaluate their potential use as biomarkers of BC and to correlate their serum levels with the clinicopathological features to detect the diagnostic potential of these miRNAs in patients with BC.Subjects and Methods : This study was conducted on 70 patients with BC and 20 apparently healthy females used as a control group. They were subjected to routine laboratory investigations and gene expression analysis using quantitative SYBR Green reverse transcription Real Time polymerase chain reaction.Results : It was found that median levels of fold change of miR-10b and miR-21 were significantly higher (up-regulated) in serum of BC patients compared to the healthy control group (p<0.001) . ROC curve analysis for miR 21 have demonstrated AUC of 0.96 [ 95% CI, 0.928 & 1 ] , with sensitivity 95.7% and specificity 85% ,and AUC of 0.99 [ 95% confidence interval (CI), 0.975&1 ], with sensitivity 97.1 % and specificity 100 % for miR 10b,Conclusions: miR21 and miR10b could be helpful biomarkers in BC diagnosis
