Twenty-two new binary and mixed-ligand transition metal complexes of cocaine and TMEDA (as a secondary ligand) with some d- and f-block transition metal elements were synthesized as new potent anticancer metal-based agents against human liver HepG-2 and breast MCF-7 cancer cells. The prepared complexes were structurally characterized by a variety of physicochemical tools such as elemental analysis, FT-IR, UV-vis, mass spectroscopy, scanning electron microscope (SEM),molar conductance, and thermal analyses techniques.Structural, spectroscopic, electronic, and thermodynamic properties were also studied theoretically by performing density functional theory (DFT) and time-dependent density functional theory (TD-DFT) calculations for the purpose of correlating between the experimental and theoretical results.The newly synthesized complexes were also investigatedfor their antiproliferative activityagainst HepG-2 and MCF-7 cancer cell lines.Further, in vitro testing was carried out to study their antimetastatic properties, apoptosis inducing characteristics, effect on tumor suppressor proteins(P21 and P27) level and effect on glutathione peroxidase (GPx) level in MCF-7 cells.In addition, the antimicrobial activities of the formed metal complexes were studied in terms of their antibacterial activities against Staphylococcus aureus and Bacillus subtilisas Gram positive bacteriaand Salmonella typhimurium and Escherichia colias Gram negative bacteria comparing to gentamycin and their antifungal activities towards Aspergillusfumigatusand Candida albicanscomparing to ketoconazole using the agar well-diffusion technique
