Sepsis remains a major problem for both scientists and clinicians. Cecal ligation and puncture (CLP) considered as the gold standard for animal models of sepsis. The aim of the present study was to assess the complications which associated with sepsis induced by CLP and the potential curative role of Sepia officinalis body tissue (SOBT) and Sepia officinalis polysaccharide (SOP) extracts in hepatorenal injury induced by CLP. Fourty eight male albino rats (12 rats/each group) were divided into four groups, the Sham-operated control, CLP, SOBT and SOP groups. Both Sham and CLP groups administered distilled water while, SOBT and SOP treated groups administered SOBT and SOP for 2 days after CLP. On the third day, half of the rats were euthanized for samples collection. But, the other half was still live and used for survival study. The present study revealed that the SOBT and SOP extracts showed in vitro bactericidal activity against Gram positive and Gram negative bacteria. Furthermore, SOBT extract and SOP treatment increase the percentage of survival rate of the rats by (66.7%) and (83.33%), respectively, as compared to the untreated CLP rats. In addition, the results revealed that CLP operation caused anemia, leukocytosis, thrombocytopenia, elevation in procalcitonin level and oxidative stress in liver and kidney of rats, which have bad impacts on liver and kidney functions and decrease in cytochrome P450 1A2 activity. However, SOBT and SOP showed their ability to improve the hematological parameters, procalcitonin level and reduce the oxidative stress; which caused significant improvement in most of the biochemical parameters. Also, histopathological examination showed an improvement in the liver and the kidney architecture of the treated groups comparable with untreated one
