These studies were conducted at Therpautical Chemistry department , Pharmaceutical and Drug Industry Division and the department of Plant Biotechnology, Genetic Engineering and Biotechnology, National Research Center, Cairo, Egypt. The present investigation was conducted to address the following objectives: Establishment of plant tissue culture system of Cleome droserifolia and the possibility of using in vitro cell cultures in the production of bioactive substances and determine the hypoglycemic, oxidative stress markers and hepatoprotective effect of methanolic crude callus extract of Cleome droserifolia in STZ induced diabetic rats and normal rats.The most important outcomes of these studies are as follows: 1- Effect of different growth regulators (auxins and cytokinins) in production of calli cultures from different explants of cleome droserifolia The frequencies of callus formation differed significantly among different combinations of growth regulators and different explants. MS media accompanied with 0.5 mg/l NAA +1 mg/l BA (M3) showed the highest mean of callus formation, then MS media supplemented with 1 mg/l BA (M5) in both hypocotyl and root explants. The maximum of callus percentage was observed with root explants, while MS media supplemented with 1 mg/l BA (M5) showed the highest mean of embryogenic callus, hypocotyl explants produced the highest callus fresh and dry weights followed by the root
