Diabetic foot ulcer is a disastrous complication of diabetes mellitus that may end up with leg amputation.MRSA strains have the ability to be resistant to most Ý-lactam antibiotics, but also to a wide range of other antimicrobials, making infections difficult to manage and very costly to treat. MRSA in diabetic foot disease is increasing worldwide to develop into a problem of healthcare provision. MRSA biofilm is a serious threat as it is considered responsible for chronic or persistent infections and may cause therapeutic failure with regular antibacterial therapy. Aim of this study : Detection of frequency of MRSA recovering among diabetic foot patient, Studying of antibiotic resistance pattern and biofilm formation for MRSA isolates and detection of presence of genes icaA and icaD responsible for biofilm production. Subjects and methods: one hundred and fifty (150) patients with persistent diabetic foot ulcers was involved in this study. Samples obtained from ulcers were directly plated on MacConkey, blood and mannitol salt agar, incubated aerobically all colonies appeared were examined macroscopically and different pathogens identified by gram stain and the available biochemical reactions Coagulase and catalase tests for identification were done. We used VITEK 2 system to confirmed identification isolated bacteria. Colonies of S. aureus were screened for resistance to methicillin on Mueller{u2013}Hinton agar supplemented with oxacillin at 4 og/mL Antibiotic sensitivity test was investigated using Kirby Bauer Disc Diffusion method.. MRSA strains examined for biofilm formation by tissue culture plate method. Detection of icaA and icaD genes was investigated by PCR
