Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Science - Department of Biochemistry Summary: In the present study, development of a testosterone radioimmunoassay (RIA) technique using liquid phase double antibody is the main objective. The production of polyclonal testosterone antibody was undertaken by immunizing seven females white New-Zealand rabbits. The preparation of radio-iodinated testosterone tracer was carried out. A set of testosterone standards was prepared using highly purified testosterone antigen. Formulation, optimization and validation of the local testosterone double antibody liquid phase RIA system were carried out.The result obtained show that the RIA system of testosterone is sensitive, specific and accurate and can be used in diagnosis of variety of diseases especially hypogonadism and possible male infertility
