Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Science - Department of Biochemistry Summary: Breast cancer is a leading cause of cancer-related deaths in women worldwide. The RECK metastasis suppressor gene encodes a membrane associated MMP regulator protein that is able to suppress tumor invasion and metastasis by negatively regulating MMPs involve in carcinogenesis, such as MMP-2. This research was performed to assess RECK ,MMP2 genes amplification by RT- PCR technology, and ER ,PR, HER-2/neu expression using Immunohistochemical (IHC) staining, these markers were studied in 50 breast female cancer patients. The results of this study were that RECK gene amplification, by RT-PCR was positive in 20% while MMP2 gene amplification was positive in 30% of invasive duct breast cancer and showed the MMP2 gene was significantly associated with bad prognostic parameters as lymph nodes involvement and absence of Estrogen and Progesterone receptors. RECK gene was significantly associated with favorable prognostic factors as absence of lymph nodes involvement and presence of Estrogen and progesterone receptors. Finally it could be concluded that RECK and MMP2 genes are a significant prognostic factor correlated with invasive breast cancer. RECK and MMP2 genes expression is therefore a potentially useful prognostic marker for breast cancer
