Dissertation note: Thesis (M.Sc.) - Cairo University - Faculty of Pharmacy - Department of Pharmaceutical Chemistry Summary: This thesis is concerned with the development of simple and accurate methods for the analysis of some drugs affecting cardiovascular system, including enalapril maleate, irbesartan, hydrochlorothiazide and nitrendipine that are commonly used for the treatment of different forms of cardiovascular disorders. The studied drugs were analyzed in drug substance, in laboratory prepared mixtures in presence of their impurities, in their pharmaceutical dosage form or in plasma. An HPLC method using diode array detection was carried out for the simultaneous determination of enalapril maleate, irbesartan, hydrochlorothiazide and nitrendipine in the presence of enalapril diketopiperazine (enalapril maleate impurity) and dehydronitrendipine (nitrendipine impurity A). Experimental design was applied by using placket-Burman and Face-centered composite design during method optimization to obtain the best resolution with the minimum experimental trials. In addition, LC-MS/MS method was developed for the simultaneous determination of enalapril maleate, hydrochlorothiazide and nitrendipine in the presence of some of their major metabolites; enalaprilat (the active form of enalapril maleate) and dehydronitrendipine (the inactive form of nitrendipine) in human plasma using felodipine as an internal standard
