Obesity is a condition in which excess body fat may put a person at the risk of developing insulin resistance and type 2 diabetes mellitus. The aim of this study was to measure the levels of leptin, adiponectin and resistin as secretory hormones from adipose tissue to discover their role as a link between obesity and insulin resistance diabetes mellitus. Forty-five subjects divided into four groups, 15 with normal weight-as control group-(group I), 15 obese subjects (group II), ten subjects from group II followed weight reduction for 2 months (group III) and 15 obese diabetics (group IV). Serum insulin, leptin, adiponectin and resistin were measured by ELISA. Lipid profile was measured by a spectrophotometric method. Also, anthropometric measurements were performed. The study showed that the obese and obese diabetic subjects have got higher serum leptin and resistin levels when compared with controls. In contrast, serum adiponectin concentration was significantly lower in obese before diet and obese diabetic subjects compared to the control group. After weight loss, there is an improvement in all these parameters. The findings from bivariate correlation analysis were further explored using multiple linear regression analysis which confirms that resistin rather than adiponectin and leptin was an important determinant of insulin resistance. From all the above work, we conclude that even modest weight loss can improve metabolic risk factors through changes in some cytokines
