520 _aBackground: Increased activity of angiotensin-converting enzyme (ACE) in the blood and cerebrospinal fluid of patients with multiple sclerosis (MS) suggested that ACE may play a role in the pathogenesis and progression of MS. Objectives: To assess the relation between ACE gene I/D polymorphism and prevalence of MS in Egyptian patients and to investigate its possible association with disease severity and progression. Also to assess the relation between TNF-Ü and MS.Subjects and Methods: This study was conducted on 60 MS patients and 50 healthy controls. Polymerase chain reaction (PCR) was applied to detect ACE gene I/D polymorphism. TNF-Ü levels were measured by ELISA.Results: There was no statistical significant difference in the distribution of ACE I/D genotypes (p = 0.115) or in the allelic frequencies (p = 0.067) between patients with MS and control subjects. Also, there was no statistical significant difference in TNF-Ü levels between MS patients and control (p=0.113). Conclusion: There is no relation between ACE I/D polymorphism and MS susceptibility and/or severity in the study population
