520 | _aBackground: Mesenchymal stem cell (MSC) based liver tissue engineering on nano-fibrous scaffold holds great promise for cell-based therapy in end- stage liver failure. Scaffolds are three-dimensional (3D) matrices that provide the initial structural support for cells to attach, proliferate, and differentiate, facilitating the formation of an extracellular matrix (ECM). Aim of the Work: In this study we examined the differentiation potential of MSCs into hepatocytes in two- and three-dimensional (2D and 3D) culture systems to improve the in vitro-hepatic differentiation of MSCs to be used in tissue-based therapies in patients with chronic liver diseases. Materials and Methods: MSCs were isolated from human umbilical cord blood (UCB). Hepatogenic differentiation was induced on 2D and 3D culture system and hepatogenic function was evaluated by immunocytochemistry, Periodic acid Schiff staining (PAS), enzyme linked immunosorbent assay (ELISA), and gene expression analysis. Differentiated cells were injected intravenously (IV) into murine model of carbon tetra chloride (CCL4) induced liver fibrosis. Animal models were divided into 3 groups, including pathological control group injected with CCL4, CCL4-induced liver fibrotic group treated with 2D differentiated cells, and CCL4-induced liver fibrotic group treated with 3D differentiated cells. Livers were stained with anti-human alpha-feto protein (AFP) and albumin immunoperoxidase staining.
