520 | _aIntroduction: The aim of this study is to investigate the possible role of sitagliptin on improving memory defects in Alzheimer's disease in rats.This was performed by comparing sitagliptin withdonepezil in both diabetic and non-diabetic models of Alzheimer'sdisease.Material and methods: Eightyadult female albino rats, matched for age and weight (between 150 and 230 g) were used for the experiment. The rats were divided into main 4 groups:Group (1) untreated rats were left to the end of the experiment.Group (2) Sitagliptin 10 mg/kg was given orally by gavage. Group (3) Alzheimer model was done by intrapertonial injection of aluminum chloride that was dissolved in water and injected to rats in a dose of 4.2mg/kg for 28 days and subdivided into 3 groups.Group (3a) after intrapertonial injection of aluminum chloride, rats were left untreated.Group (3b) sitagliptin 10 mg/kg was given orally for 2 weeks after starting aluminum chloride injection and was used for 28 days. Group (3c) donepezil 1 mg/kg was given orally 2 weeks after starting aluminum chloride injection and was used for 28 days. Group (4) diabetes mellitus was induced by a single intraperitoneal injection of freshly dissolved streptozotocin (STZ) 60 mg/kg to 18 h fasted rats. Animals with fasting blood glucose above 200 mg/dl were included in the study as diabetics and subdivided into 3 groups.Group (4a) rats were left untreated.Group (4b) sitagliptin 10 mg/kg was given orally to diabetic rats and was used for 28 days. Group (4c) donepezil 1 mg/kg was given orally to diabetic rats and was used for 28 days | ||
