520 _aBackground: Acute myeloid leukemia (AML) is a heterogenous disease with multiple established prognostic factors. JAK2 overexpression in AML was confirmed by many authors with an influence on the overall survival. Philadelphia positive AML (Ph+ AML) is a provisional entity in the recent WHO classification where discrimination from acute blastic crisis on top of chronic myeloid leukemia (CML) is still a challenge. Aims: to study JAK2 gene expression pattern in AML patients and its relation with other established prognostic variables and outcome parameters. Also, to determine the prevalence of the BCR-ABL fusion gene in AML cases with determination of the clinical and laboratory criteria of these cases. Methods: Prospective study on 175 AML patients treated at medical and pediatric oncology departments. Jak2 gene expression was evaluated by real time RT-PCR using taqman primer probe assay and BCR/ABL fusion was tested by conventional RT-PCR. All patients received current standard of care treatment for AML at NCI, Cairo University, Egypt
