520 | _aBackground: Pityriasis versicolor (PVC) is a superficial fungal infection caused by Malassezia species. A new entity of patients was described, characterized by recurrent disseminated pityriasis versicolor (RDPV) and was proven not to be infectious. Studies demonstrated the host genetic variation in immune response, particularly the role of IL-17 in antifungal immunity and skin inflammation.Sera of this entity of patients was examined for IL-17 gene polymorphism.Aim of work: To investigate IL-17A and IL-17F gene polymorphism in pityriasis versicolor patients in an attempt to reveal their role in pityriasis versicolor susceptibility. Methodology:A total of 100 patients with recurrent disseminated PVC with lesions measuring {u2265}10cm in at least one of their extensions, {u2265} 2 episodes of PVC in a year and a positive Wood{u2019}s light and/or direct mycological examination for Malassezia species, as well as100 gender and age matched controls were enrolled in this study. From each participant, 3 ml venous blood was withdrawn and put in EDTA tube for DNA extraction of IL-17A & IL-17F genes, using DNA extraction kit. Detection of single nucleotide polymorphism (SNP) occurred using real time PCR (polymerase chain reaction) with specific primers and probe. SNP was examined for both IL17A (rs2275913) and IL17F (rs763780), respectively, using the TaqMan technique. Result:A statistically significant difference was noted between patients and controls regarding the frequency of the mutant IL-17A (rs2275913) AA gene vs normal (wild) GG gene, as well as heterozygous IL-17F (rs763780) TC gene vs normal (wild) TT(p-value <0.001, respectively).In each case, the odds ratio was increased >1 indicating higher incidence of PVC in patients with mutation of IL-17A and IL-17F. A positive family history confers to the susceptibility to RDPV (p <0.001). No correlation was found between demographic and clinical data, and the IL-17 gene polymorphism. | ||
