In the current study , the DNA vaccine coding for H5 gene of HPA1 H%N1 subtype was produced and co-administrated with DNA vaccine coding for esat6 gene of Mycobacteria (pcDNA3.1/esat6) as an immunomodulatore. first, samples taken from different governorates during 2010 and 2011 were subjected to extensive virus isolation and identification one of 2011 were subjected to extensive virus isolation and identification one of the isolated feed isolate was subjected for further investigation where the full length H5 gene was amplifed and sequenced and cloned in pENTER/D/SD/Topo entry clone and homologues recombination with pDEST40 mammalian expression destination vector was done to produce the DNA vaccine pDEST40/H5
