Introduction: The Smart seal system has been claimed to expand laterally in canal walls; providing an excellent three dimensional seal. Aim: part I: An assessment of the calcium ion release and pH of MTA Fillapex sealer compared with Smart paste bio. Part II: test the adaptation using (SEM) and (TEM) of smart seal, smart seal bio and MTA Fillapex sealers.Results: Smart paste bio showed higher mean calcium ion concentration and higher mean pH than MTA Fillapex after 24 hours and 7 days. Examination of SEM and TEM revealed both gap-free and regions and gap-containing regions in canals filled with all sealer groups at all segments. Conclusion: Both MTA Fillapex and Smart paste bio sealer promote calcium ion release and possess an alkaline pH. The Three experimental sealers had similar adaptation in middle and apical segments
