Ninety male albino rats were used in this study, 12 were used as control groups and the remaining rats were used as experimental groups, diabetes was induced in the experimental groups by streptozotocin. The submandibular and parotid glands were removed and were investigated by several ways as: Haematoxylin and eosin stain. Conventional histochemical staining. Alcian blue (Ph 2.5) for acid mucopolysaccharides. Periodic acid schiff for neutral mucopolysaccharides.Histochemical examination by lectin. Electron microscopy: diabetes mellitus is accompanied with degeneration of acini in the form of fatty changes which increase with increased duration of diabetes, these fatty changes appeared by the electron microscope as lipid droplets within the secretory granules, and within the lysosomes. Degeneration of the serous acini was more severe than the mucous acini. Thick walled and obliterated blood vessels were clearly noticed and were explained by precipitation of neutral mucopolysaccharides as they appered darkly stained by periodic acid schiff in addition to precipitation of galactose as they also appeared darkly stained by lectin. The neuropathy accompanying diabetes is due to delamination of myeline sheath of myelinated nerve fibers, sequestration and hyalinization of the axons as were shown with the electron microscope
