Background : The prevalence of Diabetes mellitus in middle-income countries is considered a major health problem that needs to be addressed appropriately. A comparative evaluation of antidiabetic activities of three selected plants cultivated in Egypt namely; mango leaves, rosemary leaves and barley grains was performed to establish their potentials. Method: The plant extracts were prepared using ethanol (70%) and fractionated with petroleum ether, methylene chloride, ethyl acetate and n-butanol. The antihyperglycemic activity of the extracts, fractions and isolated Ý-glucan were tested in-vitro using Ü-glucosidase and Ü-amylase inhibitory assays. Meanwhile the in-vivo anti-hyperglycemia and antioxidant studies were carried out on the total extracts and isolated Ý-glucan in alloxan induced diabetic rats. Quality control parameters were established for the selected samples including proximate analysis, determination of macro- and micronutrients, amino acids profile and vitamins content. Phytochemical investigation of the biologically active fractions was established followed by HPLC standardization finally LC-MS profiling of the secondary metabolites of the selected drugs
