Summary, etc. This research deals with the design and synthesis of twenty three new pyrazolo[3.4-d]pyrimidine derivatives; V, VI, VIIa,b, VIIIa,b, IXa-c, Xa.b, XI, XIIa-c, XV, XVII and XVIIIa-f as CDK2 inhibitors. Preliminary molecular modeling studies were used to support the design as well as a computational ADME study was carried out on all designed compounds. The twenty-three newly synthesized compounds were tested for their in vitro anticancer activity at the National Cancer Institute, NCI, Maryland, USA against sixty cancer cell lines. Compounds VIIa, IXc, XIIc, XVIIIa and XVIIIb displayed potent and wide range anticancer activity against almost all the cancer cell lines at the single dose screening therefore these compounds were further selected for five doses screening. Compounds VIIa, IXc, XIIc and XVIIIa were tested for their CDK2 inhibitory activity using roscovitine as a reference drug where compounds VIIa and IXc were the most potent CDK2 inhibitors and were subjected to cell cycle and apoptotic effects study.

Summary, etc. يتناول هذا البحث تصميم وتخليق بعض مشتقات البيرازولوبيريميدين الثلاثة و العشرون الجديدة كمثبطات لانزيمCDK2 و تم استخدام تقنيات النمذجة الجزيئية الاولية لدعم التصميم وتم إجراء دراسة ADME الحسابية على جميع المركبات المصممة. تم اختبار المركبات الثلاثة والعشرين المُصنَّعة حديثًا لمعرفة نشاطها المضاد للسرطان في المختبر في المعهد القومي للسرطان، ميريلاند، الولايات المتحدة الأمريكية علي ستين خلية. أظهرت المركبات VIIa، IXc، XIIc، XVIIIa وXVIIIb نشاطًا قويًا وواسع النطاق مضادًا للسرطان ضد جميع خطوط الخلايا السرطانية تقريبًا في فحص تركيز واحد ، لذلك تم اختيار هذه المركبات لفحص خمس جرعات أخرى. تم اختبار مشتقات VIIa IXc, XIIc, وXVIIIa لنشاطها المثبط لانزيم CDK2 باستخدام روسكوفيتين كدواء مرجعي. تم إخضاع مركبات VIIa وIXc مثبطات لاختبار تثبيط انزيم CDK2 ودراسة دورة الخلية وتأثيرات موت الخلايا المبرمج.
