Summary, etc. Aim: This in vitro study aimed to evaluate and compare the antibacterial efficacy and biocompatibility of Ca (OH)2, CPCP and a combination of Ca (OH)2 and CPCP as root canal dressings against E. faecalis in single-rooted mandibular premolars compared to teeth with no intracanal medications.<br/>Methodology: <br/>1.The antimicrobial test<br/>A total of fifty extracted human single-canalled teeth were collected and prepared. The teeth were sterilized and 20 µ of E. faecalis bacteria was inoculated into each root canal and divided randomly into five equal groups according to the intracanal medication. G1, the canals were medicated with CPCP using a cotton pellet placed into the pulp chamber. G2, the canals were medicated with CPCP using paper point till the apical 1/3. G3, the canals were medicated with Ca (OH)2 till the apical 1/3. G4, the canals were medicated with a combination of Ca (OH)2 <br/> and CPCP till the apical 1/3. G5, the canals were kept empty without any medications (control).The resultant growth was visually quantified by counting the number of colonies forming units per milliliter of each dilution (CFUs/ml) on the agar medium.<br/>2.The biocompatibility test:<br/>Normal fibroblast cell line BHK was obtained from the National Cancer Institute of Egypt and then cultured (24-well culture for each group). Three different intracanal medications (Ca (OH)2, CPCP, a combination of Ca (OH)2 and CPCP) were added to the cultured cells. Control Group (CG), Cells were grown in a fresh medium. Cytotoxicity was evaluated by MTT reduction assay. <br/>Results: Intracanal medication with a combination of Ca (OH)2 and CPCP showed the highest anti-bacterial effect followed by intracanal medication with Ca (OH)2. Intracanal medication with CPCP using paper point, medication with CPCP inserted into the pulp chamber on a cotton pellet, respectively with no significant difference between them (p>0.05). Root canals kept with no intracanal medications showed significantly higher post-intracanal medication bacterial count than all other groups (p<0.001). According to the biocompatibility, CPCP group showed the highest cytotoxic effect followed by the combination group and Ca (OH)2 group, respectively<br/>Conclusion: Combining Ca (OH)2 and CPCP shows high antibacterial efficacy to E. faecalis and low E. faecalis count compared to each medication alone. Ca (OH)2 was biocompatible on the tissue followed by a combination of Ca (OH)2 and CPCP then CPCP, which proves that CPCP is cytotoxic. Increasing the concentration of the medication reduces the viability of tissue.

Summary, etc. هدفت هذه الدراسة المختبرية إلى تقييم ومقارنة الفعالية المضادة للبكتيريا والتوافق الحيوي للكالسيوم هيدروكسيد ومادة البارا كلوروفينول المشبعة بالكافور والمزيج من الكالسيوم هيدروكسيد ومادة الباراكلوروفينول المشبعة بالكافور كضمادات داخل القناة الجذرية الاحادية ضد بكتيريا المكورة المعوية البرازية في الضواحك السفلية أحادية الجذور مقارنة بالأسنان التي لا توجد أدوية داخل القناة. طريقة البحث: 1. اختبار مضادات الميكروبات: تم جمع وتحضير خمسين سنًا بشريًا أحادي الجذر. تم تعقيم الأسنان وحقن 20 ميكرومتر من بكتيريا المكورة المعوية البرازية في كل قناة جذر وتقسيمها عشوائياً إلى خمس مجموعات متساوية حسب الدواء داخل القناة. G1، السن المعالج بمادة الباراكلوروفينول المشبعة بالكافور باستخدام القطن الموضوعة في حجرة اللب. G2، القناة المعالجة بمادة الباراكلوروفينول المشبعة بالكافور باستخدام قطنة ورقية حتى ثلث القناة. G3، القناة المعالجة ب الكالسيوم هيدروكسيد حتى ثلث القناة. G4، القناة المعالجة بمزيج من الكالسيوم هيدروكسيد ومادة الباراكلوروفينول المشبعة بالكافور حتى ثلث القناة. G5، ظلت القنوات فارغة دون أي أدوية (كمجموعة تحكم). تم قياس النمو الناتج بصريًا عن طريق حساب عدد الوحدات المكونة للمستعمرات لكل مليلتر من كل تخفيف (CFUs / ml) على وسط أجار باستخدام العين مجردة.
