| Summary, etc. | The importance of the neuronal Na+/K+ ATPase (NKA) pump, for maintenance of ionic gradients that underlies synaptic transmission and memory functions, has been highlighted. Amyloid-beta (Aβ) interferes with NKA pump by binding between its α and β subunits, with subsequent activation of Src family kinases promoting neurodegeneration in Alzheimer’s disease (AD). This study aims to investigate ouabain’s (OUA) role in streptozotocin (STZ)-induced AD in mice through its ability to modulate NKA function at two doses levels. Mice received a single injection of STZ (3 mg/kg/ICV). Mice were treated with low dose OUA (1.5 mcg/kg/day, i.p) or high dose OUA (1.5 mg/kg/day, i.p) for 21 days. Low dose OUA alleviated memory impairment as indicated by the Morris water maze and Y-maze tests and preserved intact neurons in hippocampus. Furthermore, low dose OUA hindered STZ-induced Aβ accumulation, neuroinflammation, restored normal gene expression of NKA subunits and reduced activation of deleterious Src and Fyn kinases. It was effective also in decreasing necroptosis as well as disease-associated microglial (DAM) markers. Captivatingly, low dose OUA, but not high dose, might function as a probable neuroprotective agent against AD through endorsing NKA activity which in turn hampered Src kinases activation, neuroinflammation and tumor necrosis factor-α-dependent necroptosis. In addition, a low dose of OUA amended lysosomal impairment; thus, enhancing Aβ clearance through hindering DAM. |

| Summary, etc. | مرض الزهايمر هو الشكل الأكثر شيوعًا للخرف و يمثل مرض الزهايمر مصدر قلق صحي عالمي كبير حيث بلغ عدد الاصابات إلى حوالي 55 مليون شخص في جميع أنحاء العالم. يعد تجميع وترسب بيتا الأميلويد بالإضافة إلى فرط فسفرة تاو هي السمة المميزة لمرض الزهايمر، ويلعب دورًا حاسمًا في التسبب في المرض و تقدمه. بالاضافة إلى ذلك، يلعب الالتهاب العصبي دوراً مهمًا في التسبب في مرض الزهايمرحيث انه من المعروف أن الخلايا الدبقية، بما في ذلك الخلايا الدبقية الصغيرة والخلايا النجمية، يتم تنشيطها من خلال تراكم بروتين تاو مفرط الفسفرة وأميلويد بيتاز<br/>الجدير بالذكر ان لمضخة الصوديوم و البوتاسيوم دور كبير في تطور مرض الزهايمر. تتوزع مضخة الصوديوم و البوتاسيوم بشكل كبير بين الأنسجة المختلفة و تلعب دور حاسم في الحفاظ على التوازن الأيوني عبر غشاء الخلية. بعض الدراسات اشارت إلى دور مضخة الصوديوم و البوتاسيوم حيث يتداخل الأميلويد بيتا بين الوحدتين الالفا و البيتا مما ينتج عنه بالتبعية التنشيط لكينازات عائلة Src التي تعزز التنكس العصبي في مرض الزهايمر. |
