| Summary, etc. | Tomato Yellow Leaf Curl Virus (TYLCV) poses a significant threat to tomato production in Egypt. Visual observations, serological, and molecular techniques were employed to detect and quantify the virus in infected plants. Positive samples were categorized based on symptom severity to explore the variable effects of different TYLCV isolates. Disease Severity Index (DSI), incidence, and virus concentration were analyzed to understand the behavior and impact of these isolates. DAS-ELISA, PCR, and multiplex-PCR (mPCR) targeting the partial coat protein (pCP) gene were used for virus detection. Phylogenetic analysis revealed that the isolates were highly related (99%) to the TYLCV-IL group and moderately related (97%) to the TYLCV-mild group. Betasatellite presence and its association with symptom severity were also investigated. To control the TYLCV, two regions from the viral genome located in the overlap regions between C1/C2 (180bp), and V1/V2(240bp) each alone, and the two regions together (C1/C2+V1/V2) were used for studying their efficiency in working as siRNA to target viral DNA accumulation in plant cells. Constructs were prepared by cloning each of the two regions both in the sense/antisense directions in the binary vector pFGC5941 and Agrobacterium LB4404 was used in transient expression employing the Syringe Spotted technique in tomato plants. However, infiltered plants with the construct containing the two regions C1/C2+V1/V2 and the C1/C2 regions proved to be the best region for initiating PTGS as it inhibited the accumulation of the viral genome as well as inhibiting the appearance of viral symptoms in the tested plants than the V1/V2 construct. It can be concluded that this mechanism can result in higher viral resistance in tomatoes against TYLCV. The study highlights the genetic diversity and biological variability of TYLCV isolates in Egypt and posttranscriptional gene silencing as a virus resistance method. Understanding these characteristics is crucial for developing effective strategies to combat the Tomato Yellowing Leaf Curl Virus in tomato plants. |
