| Summary, etc. | Ticks have harmful impacts on both human and animal health and cause considerable economic losses. Leucine aminopeptidase enzymes (LAP) play important roles during tick infestation to liberate vital amino acids necessary for growth. The aim of the current study is to identify, express and characterize the LAP from the hard tick Hyalomma dromedarii, elucidate its biochemical characteristics and test its potential effect against tick infestation. An open reading frame of 1560 bp encoding a protein of 519 amino acids was identified and cloned. The LAP full-length was expressed in Escherichia coli BL21 (DE3) and purified. The recombinant enzyme (H.d rLAP- 6×His) had a predicted molecular mass of approximately 55 kDa. The purified enzyme showed maximum activity at 37°C and pH 8.0–8.5 using Leu-p-nitroanilide as a substrate. The activity of H.d rLAP- 6×His was sensitive to β-mercaptoethanol, DL-dithiothreitol, 1,10- phenanthroline, bestatin HCl, and EDTA and completely abolished by 0.05 % SDS. In parallel, the enzymatic activity was enhanced by Ni2+, Mn2+ and Mg2+, partially inhibited by Na+, Cu2+, Ca2+ and completely inhibited by Zn2+. Purified rLAP- 6xHis was used to immunize rabbits in three doses at two-week intervals. Anti- rLAP- 6xHis IgG levels increased gradually after each dose and reached their maximum 2 weeks after the second booster dose as revealed by ELISA. The specificity of rLAP- 6xHis was confirmed by western blot analysis. <br/>Also, immunization of rabbits with rLAP- 6xHis resulted in reduced blood sucking by ticks, thus providing protective immunity in the experimental model.<br/>H. dromedarii fed on rLAP- 6xHis – immunized rabbits showed reduction in their engorged weight, egg mass, egg number and reproductive index by 63.5%, 78.5%, 78.3% and 55.8%, respectively.<br/>Based on these results, rLAP- 6xHis represents a potential candidate for anti-tick vaccine against H. dromedarii infestation. |

| Summary, etc. | تتسبب الإصابة بالقراد في تأثيرات ضارة على صحة الإنسان والحيوان مما يؤدي خسائر اقتصادية كبيرة. وتلعب إنزيمات الليوسين أمينوببتيدازات أدوارًا مهمة أثناء الإصابة بالقراد لتحرير الأحماض الأمينية الحيوية اللازمة للنمو. وقد هدفت الدراسة الحالية إلى تحديد واستنساخ جين الليوسين أمينوببتيداز من قراد الهيالوما دروميدارى ، وتوضيح خصائصه الكيميائية الحيوية واختبار قدرة الليوسين أمينوببتيدازعلى الحماية من الإصابة بالقراد. تم استنساخ التسلسل الجين الذى يبلغ طوله 1560 زوجًا قاعديًا يعبر عن بروتين مكون من 519 حمضًا أمينيًا. وقد تم التعبير عن LAP بطوله الكامل في Escherichia coli BL21 (DE3) وتنقيته.<br/>كان للإنزيم المعاد الإتحاد (rLAP- 6×His) كتلة جزيئية متوقعة تبلغ حوالي 55 كيلو دالتون. كما تمت دراسة التنقية والخصائص الكيمائية للإنزيم. أظهر الإنزيم المنقى أقصى نشاط عند 37 درجة مئوية ودرجة حموضة 8.0-8.5 باستخدام المادة التفاعلية Leu- p- nitroanilide . كما كان لنشاط الإنزيم استجابة فعاليه لـ β-mercaptoethanol وDL-dithiothreitol و1,10- phenanthroline وbestatin HCl وEDTA وتم فقدان كامل لنشاطه مع 0.05٪ SDS. وبالتوازي مع ذلك، تم تعزيز النشاط الأنزيمي بواسطة النيكل و المنجنيز و الماغنسيوم ، وتم تثبيطه جزئيًا بواسطة الصوديوم و النحاس والكالسيوم وتم تثبيطه تمامًا بواسطة الزنك. كما تم استخدام هذا الإنزيم المنقى لتحصين الأرانب بثلاث جرعات بفاصل أسبوعين. وارتفعت مستويات IgG المضادة لـ rLAP- 6xHis تدريجيًا بعد كل جرعة ووصلت إلى ذروتها بعد أسبوعين من الجرعة المعززة الثالثة كما كشفت عنها ELISA. تم تأكيد خصوصية rLAP- 6xHis من خلال تحليل Western blot. كما أن تحصين الأرانب بـ rLAP- 6xHis وفر مناعة وقائية ضد الإصابة بالقراد مما أدى إلى انخفاض وزن القراد المغذى إلى 63.5٪ وكتلة البيض إلى 78.5٪ وعدد البيض إلى 78.3.٪ و55.8% من مؤشر التكاثر مقارنةً بالقراد الذي تم تغذيته على أرانب المراقبة. |
