Summary, etc. Background: The prevalence and mortality rates of colorectal cancer (CRC) are <br/>increasing at an alarming rate in recent decades. Chemoprevention strategies <br/>utilizing natural supplements provide chances to delay the CRC development <br/>and reverse the progression of CRC. Therefore, the present study was intended <br/>to develop nanoemulsion, a potential drug delivery system, for pomegranate <br/>polysaccharide (PGPs) for enhancing it’s oral bioavailability and evaluate it’s <br/>efficacy in treating CRC in rats. The current study demonstrated that PGPs <br/>successfully encapsulated within nanoemulsion with a relatively high <br/>encapsulation efficiency (92.82%) providing an effective encapsulation for <br/>delivery system in the body. The in vitro experiments showed that pomegranate <br/>polysaccharide loaded-nanoemulsion had good antioxidant and antitumor <br/>abilities more than PGPs. In vivo study demonstrated that the PGP-NE decrease <br/>body weight gain resulted from 1,2 Dimethylhydrazine (DMH) administration <br/>and help in decreasing tumor size and multiplicity, incidence and while <br/>increasing relative liver weight, colon weight length ratio and mortality. The <br/>PGP-NE act as anti-inflammatory and help to significantly reduce the increase <br/>in IL-6 and MMP9 levels which resulted after administration of DMH. The <br/>PGPs-NE significantly attenuate dyslipidemia, as indicated by significant <br/>decrease in level of total cholesterol and triglyceride. Furthermore, the PGPs-NE <br/>improve the liver function markers (AST, ALP, ALT, LDH and GGT). <br/><br/><br/>Additionally, PGPs NE attenuate oxidative stress induced by DMH, as <br/>confirmed by a significant decrease in MDA level and marked increase <br/>antioxidant markers (GSH, SOD, CAT). It was concluded that PGPs NE can be <br/>used as a promising antineoplastic remedy.

Summary, etc. الهدف:تهدف هذه الرساله الي تطوير المستحلب النانومتري من متعدد السكريات المستخلص من الرمان (PGP_NE)وتوصيفه من حيث الخصائص الفيزيائيه والكيميائيه، بالاضافه الي دراسه انشطته البيولوجيه،مثل الخصائص المضاده للاكسده، والمضاده للالتهابات، والمضاد للتكاثر الخلوي، مع تقييم سميته علي الفئران .<br/>المواد والطرق:تم تحفيز سرطان القولون في الفئران باستخدام مادة 1,2-dimethylhydrazine (DMH)، وهي مادة مسرطنة معروفة تُستخدم في النماذج الحيوانية لمحاكاة تطور سرطان القولون البشروأيضا تقييم العلامات الالتهابية والمضادة للأكسدة(مثلوMDA وGSH).<br/>النتائج:أظهرت مجموعات PGPs-NE (خاصة بجرعة 50 مجم/كجم) تحسنًا ملحوظًا في جميع المعايير مقارنة بمجموعة DMH فقط كما انخفض عدد ACF بشكل واضح وايضاارتفع مستوى المؤشرات المضادة للأكسدة وانخفضت المؤشرات الالتهاب و تم تحفيز موت الخلايا المبرمج (apoptosis) في الخلايا السرطانية من خلال تعديل تعبير الجينات المرتبطة به<br/><br/>الاستنتاج:أظهرت النتائج أن PGPs-NE أظهر نشاطًا مضادًا للسرطان أعلى بكثير من PGPs الخام وُجد أن المستحلب النانوي قد قلل من حيوية خلايا HepG2 بطريقة تعتمد على الجرعة. وكانت قيمة IC₅₀للمستحلب النانوي أقل بكثير من قيمة IC₅₀ للمستخلص الخام، مما يشير إلى فعالية أعليوأيضا تم تفسير هذا التأثير المحسّنجزئيًا بسبب الخصائص التركيبية للمستحلب النانوي، مثل الحجم النانوي وتوزيع القطر المناسب، الذي يسهل الامتصاص الخلوي ويحسن التوافر الحيوي
