| Summary, etc. | Aim: Evaluation of the antibacterial action of diclofenac potassium paste in comparison to triple antibiotic paste against Enterococcus faecalis biofilm developed in single root canals by bacterial count assessment.<br/>Methods: Forty-two human-extracted mandibular premolars with single canals were selected for this study. Teeth were prepared manually up to size 25 then decapitated to a uniform root length at 15 mm, then apices were sealed with resin composite. Teeth were sterilized in an autoclave before inoculation with E. faecalis and then incubated for 14 days at 37°C. The 1st bacteriological sample (S1) was taken after 14 days. After chemo-mechanical preparation, the 2nd bacteriological sample (S2) was taken. The specimens were divided into 3 groups (n=14) each group for testing the effect of the medicament. 1st group: (triple antibiotic paste), 2nd group; (diclofenac potassium paste), and the 3rd group (no medicament was applied) as negative control. All specimens were sealed and incubated for 7 days then the 3rd bacteriological sample (S3) was taken after medicament removal. Bacterial count assessment was done by visually counting bacterial colonies on agar plates in which the measurement unit was (CFU/ml). The significance level was set at P ≤ 0.05.<br/>\Results: There was no statistically significant difference between (TAP) group, (DP) group and (NC) group where (p=0.103) for S1 samples. There was no statistically significant difference between (TAP) group, (DP) group and (NC) group where (p=0.087) for S2 samples. There was a statistically significant difference between (TAP), (DP) and (NC) groups where (p<0.001) regarding S3 samples. A statistically significant difference was found between (NC) group and each of (TAP and DP) groups where (p<0.001). No statistically significant difference was found between (TAP) and (DP) where (p=0.098).<br/>Conclusions: Diclofenac potassium can reduce E. faecalis count with a similar antibacterial action to triple antibiotic paste. |

| Summary, etc. | غرض البحث :<br/>قامت هذه الدراسة المختبرية بتقييم التأثير المضاد للبكتيريا لمعجون ديكلوفيناك البوتاسيوم مقارنة بمعجون المضادات الحيوية الثلاثية ضد الأغشية الحيوية لبكتريا المكورات المعوية البرازية في الأسنان ذات الجذور الواحدة من خلال تقييم عدد البكتريا. <br/>طريقة البحث:<br/>تم اختيار اثنين وأربعين ضرسًا سفليًا مستخرجًا من الإنسان بقنوات فردية لهذه الدراسة. تم تحضير الأسنان يدويًا حتى مقاس 25 ثم تم قطعها الأسنان للحصول على طول جذر موحد حوالي 15 مم، وتم سد القمم بمركب الراتنج. تم تعقيم الأسنان في وعاء ضغط قبل التطعيم ببكتيريا المكورات المعوية البرازية ثم تم وضعها في الحضانة لمدة 14 يومًا عند 37 درجة مئوية. تم أخذ العينة البكتريولوجية الأولى (S1) بعد 14 يومًا. بعد التحضير الكيميائي الميكانيكي، تم أخذ العينة البكتريولوجية الثانية (العينات الثانية). تم تقسيم العينات إلى 3 مجموعات (n=14) لاختبار تأثير الدواء. المجموعة الأولى: (معجون مضاد حيوي ثلاثي)، المجموعة الثانية؛ (معجون ديكلوفيناك البوتاسيوم)، والمجموعة الثالثة (لم يتم إدخال أي دواء) كعينة تحكم سلبية. تم غلق جميع العينات ووضعها في الحضانة لمدة 7 أيام ثم تم أخذ العينة البكتريولوجية الثالثة (العينات الثالثة) بعد إزالة الدواء. تم إجراء تقييم تعداد البكتيريا عن طريق العد البصري للمستعمرات البكتيرية على أطباق الأجار حيث كانت وحدة القياس .(CFU/ml) تم ضبط مستوى الدلالة عن P ≤ 0.05 .<br/>النتائج:<br/>لم يكن هناك فرق ذو دلالة إحصائية بين مجموعة معجون المضاد الحيوي الثلاثي ومجموعة ديكلوفيناك البوتاسيوم ومجموعة المقارنة السلبية لنتائج العينات الأولى. لم يكن هناك فرق ذو دلالة إحصائية بين مجموعة معجون المضاد الحيوي الثلاثي ومجموعة معجون ديكلوفيناك البوتاسيوم ومجموعة المقارنة السلبية لنتائج العينات الثانية. كان هناك فرق ذو دلالة إحصائية بين مجموعات معجون المضاد الحيوي الثلاثي ومجموعة معجون ديكلوفيناك البوتاسيوم ومجموعة المقارنة السلبية حيث كانت فيما يتعلق بنتائج العينات الثالثة. كما وجد فرق ذو دلالة إحصائية بين مجموعة المقارنة السلبية وكل من مجموعتين معجون المضاد الحيوي الثلاثي ومعجون ديكلوفيناك البوتاسيوم ولم يوجد فرق ذو دلالة إحصائية بين معجون المضاد الحيوي الثلاثي و معجون ديكلوفيناك البوتاسيوم. <br/>النتائج المستنتجة:<br/> يمكن أن يقلل ديكلوفيناك البوتاسيوم عدد بكتريا المكورات المعوية البرازية مع تأثير مضاد للجراثيم مماثل لمعجون المضادات الحيوية الثلاثية.<br/>التوصيات:<br/>ضمن حدود هذه الدراسة ، يمكن اعتماد ديكلوفيناك البوتاسيوم كبديل لمعجون المضادات الحيوية الثلاثية كدواء داخل قنوات العصب. ويتطلب الأمر إجراء تجارب سريرية عشوائية لتقييم تأثير ديكلوفيناك البوتاسيوم على كمية السموم الداخلية وفعاليته المضادة للبكتيريا باستخدام أساليب بيولوجية جزيئية، مما يمنحها قوة إحصائية. |
