| Summary, etc. | Priestia megaterium BCGT6 was isolated from rotten potato and screened <br/>for cyclodextrin glucanotransferase (CGTase) production using Horikoshi Ⅱ <br/>medium. For enzyme production P. megaterium BCGT6 was grown in <br/>media containing soluble starch, yeast extract, peptone, sodium carbonate, <br/>K2HPO4 and MgSO4·7H2O. Optimization process showed that using <br/>ammonium chloride and rice starch (at pH 8 and 30 ⁰C) enhanced the growth <br/>of bacteria and the activity of enzymes. The partially purified β-CGTase <br/>exhibited highest activity at temperature of 50 ⁰C and pH 9 with obvious <br/>thermal stability. These findings suggested the potential use of CGTase in <br/>bioconversion of starch to cyclodextrin which can be used in food industry, <br/>pharmaceutical applications, environmental applications and textile industry. |

| Summary, etc. | تم عزل بكتيريا Priestia megaterium BCGT6 من البطاطس المتحللة وتم فحص قدرتهاعلى إنتاج انزيم السيكلوديكسترين جلوكوترانسفيريز، تمت زراعة البكتيريا في وسط يحتوي على النشا القابل للذوبان، ومستخلص الخميرة، والبيبتون، وكربونات الصوديوم، وفوسفات ثتائى البوتاسيوم هيبتاهيدراتي كبريتات المغنيسيوم. كشفت عملية التحسين أن استخدام نشا الأرز وكلوريد الأمونيوم عند درجة الحموضة 8 و 30 درجة مئوية لمدة 73 ساعة أدى إلى تحسين نمو البكتيريا ونشاط الإنزيم. أظهر إنزيم β-CGTase المنقى جزئياً أعلى نشاط عند درجة حرارة 50 درجة مئوية ودرجة الحموضة 9. أظهر الإنزيم ثبات حراري جيد عند درجات حرارة عالية حيث أنه مستقر تماماً عند درجة حرارة 50 درجة مئوية. تشير هذه النتائج إلى إمكانية استخدام CGTase في التحويل الحيوي للنشا إلى سيكلوديكسترين والذي يمكن استخدامه في الكثير من التطبيقات الصناعية |
