Ionizing radiation has harmful effects on living things, and humans are exposed to far more of it now that technology is advancing so quickly. Researchers investigated the radioprotective properties of many compounds, such as aminothiols. Amifostine is the only radioprotector that was clinically approved to palliate side effects in patients undergoing radiotherapy. Amifostine has a good protective effect but is relatively toxic and has some inappropriate effects such as nausea, vomiting, and hypotension. Hence, it is required to search for less toxic and more potent radioprotectors which can be easily administered. The goal of this study is to determine whether giving the medicinal herbs quercetin and curcumin, which are well-known for their antioxidant properties, to rats can protect them from damage caused by whole-body gamma irradiation (4 times 2Gy/every 3 days up to 8Gy) both prophylactically and therapeutically.<br/> Male albino rats weighing 120–150g were split up into six groups, each including 7 rats, for the current investigation. Group 1 was the control group and received distilled water treatment. For four weeks, Group 2 was administered a daily oral dose of Quercetin (1.25 g/kg). For four weeks, Group 3 was administered a daily oral dose of 100 mg/kg of curcumin. Group 4 received 8Gy of γ-irradiation throughout their body (4 times 2Gy/every 3 days up to 8Gy). Groups 5 and 6 were administered 1.25 g/kg and 100 mg/kg of quercetin and curcumin, respectively, for 4 weeks prior to and following γ-irradiation. Following the completion of either distilled water or Quercetin and curcumin for one, two, or three weeks, six rats from each group were sacrificed. Rats exposed to whole body gamma irradiation showed suppressed blood glutathione content (GSH), superoxide dismutase (SOD), and catalase (CAT) activities, white blood cells (WBCs) and platelets (PLT), as well as increased plasma levels of alanine transferase (ALT), aspartate transferase (AST), Alkaline phosphates (ALP), total protein, albumin, urea, creatinine, uric acid, lipid profile, and lipid peroxidation (TBARS). The following were elevated: mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), mean corpuscular volume (MCV), hematocrit value (Hct%), red blood cells (RBCs), hemoglobin content (HB), and mean corpuscular hemoglobin (MCH). Furthermore, radiation caused cellular, fibrotic, and inflammatory damage to the kidney and liver. Administration of Quercetin and curcumin significantly decreased the elevation of (TBARS) and the decline in antioxidant parameters induced by irradiation as well as amelioration of liver and kidney function and blood count associated with amelioration of the histological changes induced by gamma irradiation. <br/> Conclusion: This study demonstrates that both curcumin and quercetin exhibit significant protective effects against radiation-induced hematological, biochemical, and histological alterations in male albino rats. The administration of these phytochemicals mitigated radiation-associated damage, as evidenced by restoration of erythrocyte, leukocyte, and platelet counts to near-normal levels, alongside the stabilization of biochemical markers such as reduced oxidative stress indicators (MDA, SOD, CAT) and improved liver/kidney function markers (ALT, AST, creatinine). Histopathological analysis further confirmed their protective role. Notably, the combination of curcumin and quercetin exerted a more pronounced radioprotective effect than either compound alone, suggesting potential synergism in their mechanisms of action. This enhancement may be attributed to their complementary antioxidant properties, with curcumin scavenging free radicals and quercetin upregulating endogenous defense systems. <br/>In summary, this work underscores the therapeutic potential of curcumin and quercetin as natural radioprotectors, offering a viable strategy to counteract radiation-induced damage. |

تهدف هذه الدراسة الى تقييم الدور الوقائي المحتمل للكويرسيتين والكركمين في تحسين الأضرار التي يحدثها التعرض للإشعاع الجامي وذلك من خلال دراسة التغيرات البيوكيميائية والبيوفيزيائية و الهستولوجية التي تحدث في نشاط بعض الإنزيمات. كما تم قياس التغيرات التي تحدث في مستوى مضادات الأكسدة الداخلية ومحتوى الدهون فوق المؤكسدة ومستوى الجلوتاثيون وبعض المعايير الهيماتولوجية (كرات الدم البيضاء والحمراء والهيموجلوبين وخلايا الكبد والكلى) للجرذان بعد تعريض جسمها الكلي لجرعة إشعاعية مجزأة قدرها 8 جراي.<br/>أظهرت نتائج هذه الدراسة أن تعرض الجسم الكلي لأشعة جاما يؤدي إلى زيادة ملحوظة في معظم المعايير البيوكيميائية وخلل في نشاط بعض الإنزيمات التي تحت الدراسة. وتكون هذه الزيادة مصحوبة بنقص ملحوظ في مستوى كل من الجلوتاثيون ومستوى نشاط إنزيمات مضادات الأكسدة وذلك في معظم أوقات التجربة. كما أن حقن الجرذان بمستخلص الكويرسيتين والكوركومين عن طريق الفم في الجرذان يومياً بجرعة قدرها (1.25 مليجرام/ك.جرام وزن الجسم) (100ميكرو جرام/ ك.جرام وزن الجسم) لمدة 28 يوم قبل التعرض للإشعاع يقلل من التغيرات البيوكيميائية الضارة التي تحدث في خلايا الكبد والكلي للجرذان المعرضة للإشعاع. |
