Summary, etc. Background: Paediatric immune thrombocytopenia (ITP) is an autoimmune condition whose cause is unclear and appears to include a number of different factors.<br/>Objectives: Evaluation of the potential value of the long non-coding RNA TCONS_00003870 and its targeted genes; hsa-let-7a-5p and SEMA4C genes in patients with idiopathic thrombocytopenic Purpura (ITP).<br/>Methods: In this investigation, quantitative real-time PCR was used to examine the expression profiles of each of the long non-coding RNA TCONS_00003870, the miRNA Let-7a-5p and mRNA SEMA4C in 70 Plasma samples from children with ITP and 30 from healthy controls (HCs). It was also examined whether they were associated with ITP clinical characteristic parameters and whether it's potential usage as a non-invasive circulating biomarker for ITP diagnosis.<br/>Results: Children with ITP had significantly decreased level of both lncRNA TCONS and m-RNA SEMA4C (expressed as fold changes) compared with controls (P < 0.00).When compared to HCs, ITP patients' levels of the miRNA Let-7a-5p were considerably higher (P < 0.0001). The two biomarkers miRNA Let-7a-5p, m- RNA SEMA4C had an excellent sensitivity (100%), for TCONS, it had a great sensitivity (98.6%).The all three markers have great specificity (87.1, 96.8 and 90.3<br/>% for TCONS, Let-7a-5p and the SEMA4C, respectively).

Summary, etc. نقص الصفيحات المناعي (ITP) هو مرض مناعي ذاتي يتميز بتعداد منخفض للصفائح الدموية وزيادة خطر النزيف. يظل الحدث الأولي الذي يؤدي إلى المناعة الذاتية للصفائح الدموية غير واضح ، ولكن يوجد دليل قوي على أن العديد من عوامل الخطر الجيني )مثل التعددية في الجينات المرتبطة بالحصانة( تهيئ لـ .ITP<br/>الهدف من الدراسة: تقييم القيمة المحتملة للجينات TCONS_00003870 RNA الطويلة غير المشفرة وجيناتها المستهدفة "جينات hsa-let-7a-5p و "SEMA4C في المرضى الذين يعانون من فرفرية نقص الصفيحات الدموية مجهول السبب .(ITP)<br/>طريقة العمل: تم قياس قيمة الجينات TCONS_00003870 RNA الطويلة غير المشفرة وجيناتها<br/>المستهدفة "جينات hsa-let-7a-5p و SEMA4C فى بلازما الدم باستخدام تقنية PCR وتم تقسيم<br/>المجموعات محل الدراسة الى مجموعتين :مجموعة )أ( تتكون من 30 شخص من الاطفال الأصحاء والمجموعة )ب( تتكون من 70 شخص مصاب بنقص الصفائح الدموية مجهول السبب والتى تم تقسيمها<br/>الى مجموعتين مجموعات فرعية تبعا لشدة نقص الصفيحات الدموية )حالات نقص الصفيحات الخفيف والمتوسط.(<br/>القياسات والاستنتاج: انخفضت معدلات كلا من TCONS وSEMA4C فى بلازما الدم لمرضى نقص<br/>الصفيحات المناعي (ITP) وارتفعت معدلات let-7a-5p فى بلازما الدم لمرضى نقص الصفيحات<br/>المناعي (ITP) الذى أثبت فى هذه الدراسة انه كل منهم لديه حساسية شديدة للمرض.
