| Summary, etc. | Cranial cruciate ligament (CrCL) rupture is a common orthopedic disorder in dogs, leading to stifle joint instability and osteoarthritis. This thesis aimed to develop and evaluate innovative intra-capsular CrCL reconstruction systems and introduce alternative educational models for veterinary orthopedic training. The study was conducted in three phases: cadaveric, experimental, and educational phase. The cadaveric study involved 21 stifles to assess the characteristics of the native CrCL, develop two reconstruction systems (3D-braided silk-polyester (S-PET) and 3D-printed thermoplastic polyurethane (TPU)), and evaluate the biomechanics of 14 reconstructed stifles. Mechanical testing showed that the S-PET system demonstrated significantly higher post-reconstruction tensile strength (952.6 N) than the native CrCL (518.4 N) and TPU graft (336.2 N). However, the TPU graft exhibited greater elongation (170%) compared to the native CrCL (49.3%) and the S-PET graft (90%). Both systems effectively restored joint stability after experimental CrCL excision. In the experimental study, six dogs were divided into two groups, with each group receiving one of the reconstruction systems. Over six months, all dogs regained normal limb function within one month. Both systems showed strong potential for stifle stabilization with minimal cartilage erosion and no signs of osteoarthritis. The S-PET-collagen supported tissue integration and early remodeling, combined with partial degradation. In contrast, the TPU graft offered superior mechanical stability, but its slow degradation limited biological integration and remodeling. The educational study developed 3D-printed stifle models and an Elnady plastinated canine limb specimen. The 3D models accurately represented normal and osteoarthritic joints, serving as effective tools for teaching stifle anatomy and pathology. The Elnady technique produced an odorless, durable, and anatomically detailed specimen, except for limited flexibility. Modifications in the plastination process were recommended to improve the specimen's quality for surgical training. In conclusion, intra-articular stabilization of CrCL-deficient stifles using either the S-PET-collagen or the 3D-printed TPU reconstruction systems could be a promising technique for CrCL-deficient canine stifles. The 3D-printed stifle models and Elnady plastinated canine limb specimens can serve as long-term educational tools for teaching veterinary students basic anatomy and surgical approaches. |

| Summary, etc. | يلعب الرباط الصليبي الأمامي دورا حيويا في تثبيت مفصل الركبة في الكلاب ، حيث يمنع الدفع الأمامي والدوران الداخلي لعظمة الساق ، وفرط التمدد للمفصل. تمزق الرباط الصليبي الأمامي هو سبب شائع لعرج أطراف الحوض في الكلاب ، مما يؤدي إلى الألم والعرج وتطور التهابات المفصل. يتمتع الرباط الصليبي الامامي الممزق بقدرة ضعيفة على الشفاء من تلقاء نفسه ، حتى عندما يتم خياطة الأطراف الممزقة ، مما يجعل التدخل الجراحي الخيار المثالي لاستعادة وظيفة المفاصل ، وتثبيت المفصل ، وتأخير تطور التهابات المفصل.<br/>هناك العديد من التقنيات الجراحية ، من ضمنها التقنيات القائمة على قطع العظم مثل TPLO و TTA و CTW والتي تعمل بشكل فعال على استقرار المفصل عن طريق تغيير الميكانيكا الحيوية للمفصل بدلا من استبدال الرباط الصليبي الامامي ، ولكنها مرتبطة بأوقات الشفاء الطويلة والمضاعفات الجراحية والتكاليف المرتفعة. يوفر التثبيت خارج المحفظة بديلا أبسط ، ولكنه يستخدم بشكل عام في الكلاب الأقل نشاطا أو الأصغر حجما نظرا لانخفاض فعاليته على المدى الطويل. تعتبر إعادة بناء الرباط الصليبي الامامي داخل المفصل معيارا اساسيا في إصلاح الرباط الصليبي الأمامي في الانسان، ولكنها واجهت تحديات في الكلاب بسبب صعوبات التثبيت ومشاكل الاندماج البيولوجي. وبالتالي ، فإن الدراسة الحالية كانت من ضمن دوافعها التطورات الحديثة في هندسة الأنسجة ، والتي تقدم حلولا واعدة لتحسين أداء الاربطة الصناعيه ، وتعزيز تجديد الأربطة و النتائج الاكلينيكيه.<br/>تهدف الدراسة الحالية إلى إنشاء وتقييم نظامين مبتكرين لإعادة البناء داخل المفصل لتثبيت الركبة التي تعاني من قطع في الرباط الصليبي الامامي ، باستخدام نهج انتقالي من مرحلتين (مراحل التجارب علي عينات رجول ثم التجارب علي كلا حية). تم تطوير المرحلة الأولية (دراسة عينات الرجول) لتصميم وإنشاء أنظمة إعادة البناء ، وتقييم خصائصها الميكانيكية الحيوية قبل وبعد التثبيت الجراحي ، وتحديد التقنية الجراحية وطريقة التثبيت للتطبيق التجريبي والاكلينيكي اللاحق. |
