| Summary, etc. | Foot-and-mouth disease virus (FMDV) remains a most important risk to livestock worldwide, complicated by its serotype diversity and rapid mutation rate. This thesis aimed to fast-track FMD vaccine development through two complementary approaches. First, an immunoinformatic pipeline was applied to design a heptavalent multiepitope vaccine targeting circulating Egyptian FMDV strains, with epitopes showing high antigenicity, stability, and strong binding to immune receptors. Second, flexible recombinant constructs for FMDV serotype O structural proteins (VP0, VP1, VP3) were developed using optimized degenerate primers and validated for high expression in E. coli. Results demonstrated promising in-silico vaccine properties and a scalable protein production platform, supporting future diagnostics and tailored vaccine production. |

| Summary, etc. | فيروس الحمى القلاعية (FMDV) لا يزال يشكل أحد أهم المخاطر التي تهدد الثروة الحيوانية على مستوى العالم، نتيجة لتنوع أنماطه المصليّة وسرعة تحوّره. هدفت هذه الرسالة إلى تسريع تطوير لقاحات الحمى القلاعية من خلال منهجين تكميليين. أولاً، تم تطبيق مسار معلوماتي مناعي (immunoinformatic pipeline) لتصميم لقاح متعدد الحلقات (heptavalent multiepitope vaccine) يستهدف السلالات المنتشرة لفيروس الحمى القلاعية في مصر، حيث أظهرت الحلقات المنتخبة مستويات عالية من المناعية والثبات وقدرة قوية على الارتباط بمستقبلات الجهاز المناعي. ثانيًا، تم تطوير تراكيب وراثية مرنة للبروتينات البنيوية (VP0, VP1, VP3)لفيروس الحمى القلاعية من النمط المصليO باستخدام بادئات منزوعة التخصص (optimized degenerate primers) وتم التحقق من كفاءتها في التعبير العالي داخل بكتيريا الإيشيريشيا كولاي (E. coli). أظهرت النتائج خصائص واعدة للقاح مصمم حاسوبيًا، بالإضافة إلى منصة إنتاج بروتيني قابلة للتوسع، مما يدعم تطبيقات التشخيص وإنتاج لقاحات مخصصة مستقبلاً. |
