| Summary, etc. | One of the causes that leads to the accumulation of cadmium (Cd) in soil is the excessive use of chemical fertilizers and pesticides in agricultural practices, which has a detrimental effect on global food security and safety. Root-associated bacteria, such as those that promote plant growth, have been employed as an efficient and sustainable method of detoxifying and lowering the toxicity of Cd in soil. In this work, four isolates that exhibit Cd tolerance and possess plant growth-promoting traits (phosphate solubilization, nitrogen fixation, and indole acetic acid synthesis) were identified by 16S rRNA as Serratia rubidaea. By sequencing the genome of the most promising isolate, S. rubidaea SR19, genes associated with heavy metal tolerance, stress defense, and plant growth promotion were identified. To investigate how SR19 mitigates Cd stress in cucumber plants, seedlings were subjected to different concentrations of CdCl2 for 7 days in the hydroponic system. It was found that SR19 promoted the growth of shoots, roots, and lateral roots under Cd toxicity. Moreover, reducing oxidative stress and improving overall stress tolerance in SR19-treated seedlings was observed by boosting glutathione and peroxidase activity and decreasing MDA levels. On the other hand, RNA-seq analysis identified the genes differentially expressed in the SR19-primed seedlings under Cd stress and revealed upregulation of stress-responsive genes related to metal detoxification and antioxidant defense pathways. Our research may further our knowledge of the complicated molecular processes that control how plants and PGPR interact under cadmium stress. |

| Summary, etc. | يعتبر تراكم عنصر الكادميوم (Cd) في التربة من أسباب الإفراط في استخدام الأسمدة والمبيدات الحشرية الكيميائية في الممارسات الزراعية، مما يؤثر سلبًا على الأمن الغذائي العالمي وسلامته. وقد استُخدمت البكتيريا المرتبطة بالجذور، كتلك التي تُعزز نمو النباتات، كطريقة فعّالة ومستدامة لإزالة سمية الكادميوم من التربة وخفضها. في هذا البحث، تم تحديد أربع عزلات تُظهر قدرة على تحمل الكادميوم وتمتلك صفات مُعززة لنمو النبات (إذابة الفوسفات، تثبيت النيتروجين، وتخليق حمض الإندول الأسيتيك) بواسطة الحمض النووي الريبوزي 16S، وهي Serratia rubidaea. ومن خلال تسلسل جينوم العزلة الأكثر واعدة، S. rubidaea SR19، تم تحديد جينات تحمل المعادن الثقيلة، والدفاع ضد الإجهاد، وجينات مُعززة لنمو النبات. لدراسة كيفية تخفيف SR19 لإجهاد الكادميوم في الخيار، خضعت الشتلات Cucumis sativus لتركيزات مختلفة من CdCl2 لمدة 7 أيام في نظام الزراعة المائية. عززت SR19 نمو البراعم والجذور والجذور الجانبية تحت تأثير الكادميوم. زيادة على ذلك لوحظ انخفاض في الإجهاد التأكسدي وتحسين في تحمل الإجهاد الكلي في الشتلات المعالجة بـ SR19 من خلال تعزيز نشاط الجلوتاثيون والبيروكسيديز وخفض مستويات MDA. ومن ناحية اخرى حدد تحليل RNA-seq الجينات التي تم التعبير عنها بشكل مختلف في الشتلات المعالجة بـ SR19 تحت تأثير الكادميوم، وكشف عن زيادة في الجينات المستجيبة للإجهاد والمتعلقة بإزالة السموم المعدنية ومسارات الدفاع المضادة للأكسدة. بشكل عام، قد يُعزز بحثنا معرفة العمليات الجزيئية المعقدة التي تتحكم في كيفية تفاعل النباتات ومستقبلات PGPR تحت تأثير الكادميوم. |
