| Summary, etc. | This study was designed into 3 main experiments. In these experiments full term 3 pregnant <br/>buffalo were used as amniotic membrane donor. The preparation steps of amniotic <br/>membranes were performed then preserved in refrigerator at 4°C for one week. <br/>Experiment I: 6 apparently healthy male donkeys which were equally divided into two groups <br/>(vicryl sutured and prolene sutured groups) (3 in each group), Donkeys in both groups were <br/>subjected to superficial digital flexor tendon full-thickness tenotomy at level of mid <br/>metacarpal region then treated with suturing with vicryl (size 2) in the vicryl sutured group <br/>and suturing with prolene (size 2) in the prolene sutured group using modified Kessler <br/>technique in both groups. The contralateral limb in each donkey was used as the negative <br/>control group. <br/>Experiment II: 12 apparently healthy male donkeys which classified into two equal groups (6 <br/>in each group). Donkeys in both groups were subjected to superficial digital flexor tendon <br/>full-thickness tenotomy at level of mid metacarpal region. Donkeys in amniotic membrane <br/>(AM)group were treated with suturing with vicryl (size 2) and wrapped with AM that act as <br/>scaffold and sutured by (simple interrupted) method, while control positive group donkeys <br/>were treated with suturing only (modified Kessler technique). Contralateral limb in each <br/>donkey was used as control negative group and the tendon healing evaluated by <br/>ultrasonography and MRI <br/>Experiment III: 12 apparently healthy male donkeys which divided into four equal groups (3 <br/>in each group). Donkeys in all groups were subjected to superficial digital flexor tendon full-<br/>thickness tenotomy at level of mid metacarpal region. Donkeys in amniotic membrane <br/>(AM)group were treated with suturing with Vicryl (size 2) and wrapped with AM that act as <br/>scaffold, while in the PRP (platelet rich plasma) group donkeys were treated with suturing <br/>with intralesional PRP infiltration. In group (AM & PRP group) donkeys were treated with <br/>suturing, AM as a scaffold and with intralesional PRP infiltration. In control positive group <br/>donkeys were treated with suturing only (modified Kessler technique). Contralateral limb in <br/>each donkey was used as control negative group and the tendon healing evaluated by <br/>molecular analysis, histopathology, and gross evaluation. |

| Summary, etc. | أُجريت هذه الدراسة تجريبيًا على 30 حمارًا، استقبلتها كلية الطب البيطري بجامعة القاهرة، بهدف إثبات أن الغشاء الأمنيوسي للجاموس مادة متجددة، تعمل كسقالة، ومصدر لعوامل النمو، والخلايا الجذعية التي تساعد في التئام تمزق وتر المثنية السطحي لدى الحمير دون التصاق.<br/>صُممت هذه الدراسة في ثلاث تجارب رئيسية. في هذه التجارب، استُخدمت ثلاث جاموسات عشار في تمام نموها كمتبرعة بغشاء أمنيوسي. نفّذت خطوات تحضير الغشاء الأمنيوسي، ثم حُفظ في الثلاجة عند درجة حرارة 4°م. <br/>التجربة الأولى، ستة من ذكور الحمير سليمه ظاهريا، قُسِّمت بالتساوي إلى مجموعتين مجموعة خياطة بالفيكريل و مجوعة خياطة بالبرولين (ثلاثة في كل مجموعة). خضعت الحمير في كلتا المجموعتين لقطع كامل السمك في وتر المثنية السطحي عند مستوى منطقة منتصف عظم مشط اليد الأمامية الشمال لدى الحمير، ثم عولجت بالخياطة بالفيكريل (الحجم 2) في مجموعة خياطة بالفيكريل والخياطة بالبرولين (الحجم 2) في مجموعة خياطة بالبرولين باستخدام تقنية كيسلر المعدلة في كلتا المجموعتين. استُخدم الطرف المقابل في كل حمار كمجموعة ضابطة سلبية. <br/>التجربة الثانية، اثنا عشر ذكرًا من الحمير سليمة ظاهريا ، قُسِّمت إلى مجموعتين متساويتين (6 في كل مجموعة). خضعت الحمير في كلتا المجموعتين لقطع كامل السمك في وتر المثنية السطحي عند مستوى منطقة منتصف عظم مشط اليد الأمامية الشمال لدى الحمير. عولجت الحمير في مجموعة الغشاء الأمنيوسي (AM) بخياطة باستخدام فيكريل (مقاس 2)، ولُفّت بـالغشاء الأمنيوسي الذي يعمل كسقالة، وخيطت (بطريقة بسيطة متقطعة) باستخدام فيكريل، بينما عولجت الحمير في مجموعة الضبط الإيجابية بالخياطة فقط (بتقنية كيسلر المعدلة). استُخدم الطرف المقابل في كل حمار كمجموعة ضبط سلبية، وتم تقييم التئام الوتر باستخدام الموجات فوق الصوتية والتصوير بالرنين المغناطيسي. <br/>التجربة الثالثة، اثنا عشر ذكرًا من الحمير التي تبدو سليمًه ظاهريًا، قُسِّمت إلى أربع مجموعات متساوية (3 في كل مجموعة). خضعت الحمير في جميع المجموعات لقطع كامل السُمك في وتر المثنية السطحي عند مستوى منطقة منتصف عظم مشط اليد الأمامية الشمال لدى الحمير. عولجت الحمير في مجموعة الغشاء الأمنيوسي (AM) بالخياطة باستخدام فيكريل (الحجم 2) ولُفّت بـالغشاء الأمنيوسي الذي يعمل كسقالة ، بينما عولجت الحمير في مجموعة البلازما الغنية بالصفائح الدموية (PRP) بالخياطة مع حقنPRP داخل المنطقة المصابة. في المجموعة (AM وPRP) عولجت الحمير بالخياطة، ولُفّت بـالغشاء الأمنيوسي الذي يعمل كسقالة، وتم حقن PRP داخل المنطقة المصابة. في المجموعة الإيجابية الضابطة، عولجت الحمير بالخياطة فقط (بتقنية كيسلر المعدلة). استُخدم الطرف المقابل في كل حمار كمجموعة سلبية ضابطة، وتم تقييم التئام الوتر من خلال التحليل الجزيئي والفحص النسيجي والتقييم الظاهري للوتر |
