Summary, etc. Background: Ewing sarcoma (ES) is the second most common <br/>malignant bone tumor in children and adolescents. Tumor-associated <br/>macrophages (TAMs) are a key component of the tumor <br/>microenvironment, play a central role in promoting tumor progression, <br/>metastasis and resistance to chemotherapy and radiotherapy. While <br/>increased TAM infiltration is linked to poor prognosis in various solid <br/>tumors, limited data are available regarding their role in sarcomas. <br/>Purpose: This retrospective study aimed to evaluate the frequency, <br/>density and prognostic impact of TAMs in pediatric patients with Ewing <br/>sarcoma treated at Pediatric Oncology Department, National Cancer <br/>Institute, Cairo University. <br/>Methods: Immunohistochemical analysis of CD68 and CD163 <br/>expression was performed on 74 tumor tissue samples from pediatric ES <br/>patients to assess TAM density. <br/>Results: The study cohort comprised 74 patients with a median age of 12 <br/>years (range: 0.5–18 years) and a male-to-female ratio of 0.8:1. The 5-<br/>year overall survival (OS) and event-free survival (EFS) rates were 48% <br/>and 42.8%, respectively. CD68 expression showed significant <br/>correlations with age at diagnosis (p = 0.03) and tumor size (p = 0.035). <br/>Near-significant associations were observed between CD68 levels and <br/>skeletal tumor location (p = 0.052), as well as complete response <br/>following induction chemotherapy (p = 0.054). CD163 expression was <br/>significantly associated with age at diagnosis (p = 0.04). A fair positive <br/>correlation was observed between CD68 and CD163 expression. <br/>However, neither CD68 (≤60 vs >60) nor CD163 (≤90 vs >90) levels <br/>showed significant correlation with survival outcomes (OS or EFS). <br/>Prognostic factors such as tumor site, stage, metastatic burden, response <br/>to induction chemotherapy, surgical feasibility, and histological response <br/>significantly influenced survival. Chemotherapy dose intensity (biweekly <br/>administration) showed a near-significant association with EFS (p = <br/>0.06). <br/>Conclusions: This study demonstrated relatively low survival outcomes <br/>among pediatric ES patients, consistent with data from low- and middle-<br/>income countries, and inferior to those reported in high-income settings. <br/>TAM markers (CD68, CD163) did not demonstrate a significant <br/>prognostic impact on survival. Larger-scale studies are warranted to more <br/>precisely determine the role of TAMs in pediatric Ewing sarcoma

Summary, etc. الساركوما الإيوينغ هي ثاني أكثر الأورام الخبيثة شيوعًا في العظام لدى الأطفال والمراهقين. وتعد الخلايا الضامة المرتبطة بالأورام (TAMs) من المكونات الرئيسية للخلايا المناعية المتسللة إلى الورم في بيئة الورم الدقيقة، ولها دور رئيسي كمنظمين للالتهاب المرتبط بالأورام. طُرق البحث: دراسة استعادية شملت 74 مريضًا مصابًا بساركوما الإيوينغ، تم مراجعة سجلاتهم الطبية وفحص الأنسجة الخاصة بهم. تم علاج المرضى في المعهد القومي للأورام، جامعة القاهرة، خلال الفترة من 1 يناير 2010 حتى نهاية ديسمبر 2020 النتائج: خلال فترة الدراسة، توفي 34 مريضًا. كانت فترة المتابعة المتوسطة 34.4 شهرًا (تتراوح من 0.3 إلى 107 أشهر). كانت مدة البقاء على قيد الحياة العامة المتوسطة 60 شهرًا، بينما كانت نسبة البقاء على قيد الحياة العامة التراكمية بعد 3 سنوات %55.7، وفي نهاية الدراسة كانت %48. كانت نسبة البقاء على قيد الحياة دون حدث (EFS) التراكمية بعد 3 سنوات وفي نهاية الدراسة %50.6 و%42.8 على التوالي الخاتمة: وُجدت علاقة ذات دلالة إحصائية بين تعبير CD68 وكلاً من العمر وحجم الورم الأولي. بينما يظهر تعبير CD163 علاقة ذات دلالة إحصائية مع العمر فقط. هناك ارتباط إيجابي معتدل بين CD68 وCD163. لم يكن لعلامات TAMs (CD68، CD163) تأثير تنبؤي على البقاء على قيد الحياة من حيث البقاء على قيد الحياة العامة (OS) والبقاء دون حدث (EFS)…
