| Summary, etc. | Burn wounds are a major global health concern, often complicated by bacterial infections that delay healing and increase morbidity. Cefdinir, a third-generation cephalosporin, has limited water solubility, bioavailability, and a short half-life. This study developed surfactant-based Cefdinir-loaded chitosan nanoparticles (CFD-CSNPs) to enhance topical delivery and burn healing efficacy. Using Box–Behnken Design, formulations were optimized for particle size, zeta potential, polydispersity index, and entrapment efficiency (57.89% ± 1.66). Prepared via ionic gelation, the optimized nanoparticles were spherical, showed a biphasic release (75.5% ± 3.8 in 48 h), and demonstrated superior antimicrobial, biofilm-preventive, and wound-healing effects in vitro and in vivo.<br/>Building on this, a hybrid nanofiber (NFs) system was developed by incorporating Fucoidan-coated CFD-CSNPs into polyvinyl alcohol via electrospinning. The resulting fibers were smooth, bead-free (295.12 ± 48.52 nm), exhibited optimal swelling (250.41% ± 9.59), controlled degradation (48.55% ± 1.44), and sustained release (84.00% ± 2.23 in 48 h). They enhanced skin permeation, deposition, and antimicrobial activity, promoted fibroblast proliferation, and accelerated wound closure in a murine burn model with improved tissue regeneration, TGF-β1 expression, and collagen organization. This multifunctional system offers structural support, controlled drug delivery, and the therapeutic benefits of Fucoidan, representing a promising antibacterial burn wound dressing. |

| Summary, etc. | تُعد الحروق من أبرز المشكلات الصحية عالميًا، وتزداد خطورتها بسبب العدوى البكتيرية التي تعيق الشفاء. السيفدينير، وهو مضاد حيوي من الجيل الثالث للسيفالوسبورينات، يعاني من ضعف الذوبانية وقلة التوافر الحيوي وقصر عمره الحيوي. هدفت هذه الدراسة إلى تطوير جسيمات نانوية من الكيتوسان محمّلة بالسيفدينير ومدعمة بعامل خافض للتوتر السطحي (CFD-CSNPs) لتعزيز التوصيل الموضعي وكفاءة العلاج. تم تحسين التركيبة باستخدام تصميم Box–Behnken، وأظهرت الجسيمات المحضّرة خصائص فيزيائية مثالية، وكفاءة احتجاز بلغت %57.89 ± 1.66، وشكلًا كرويًا منتظمًا، وإطلاقًا دوائيًا ثنائي الطور (%75.5 ± 3.8 خلال 48 ساعة). أظهرت التركيبة فعالية مضادة للميكروبات، وقدرة عالية على منع وإزالة الأغشية الحيوية، وتسريع شفاء الحروق في الاختبارات المخبرية والحيوانية.<br/>في المرحلة الثانية، تم تطوير نظام هجين للألياف النانوية يجمع بين جسيمات السيفدينير النانوية المغطاة بالفيكوديان وكحول البولي فينيل (Fu-cCFD-CSNPs-PVA NFs) باستخدام الغزل الكهربائي. أظهرت الألياف شكلًا متجانسًا وقطرًا متوسطًا 295.12 ± 48.52 نانومتر، وانتفاخًا مثاليًا %250.41 ± 9.59، وتحللًا محكومًا %48.55 ± 1.44، مع إطلاق دوائي مستدام (%84.00 ± 2.23 خلال 48 ساعة). كما حسّنت النفاذية الجلدية والترسيب والفعالية المضادة للميكروبات، وعززت تكاثر الخلايا الليفية وإغلاق الجروح، مع تجدد نسيجي منظم وزيادة تعبير TGF-β1. يمثل هذا النظام نهجًا واعدًا كضماد مضاد للبكتيريا لعلاج الحروق. |
