Summary, etc. To produce more effective anticancer medicines, a variety of new 2-arylideneindane-1,3-dione scaffolds were developed and synthesized by molecular hybridization. The base-catalyzed Knoevenagel reaction of 2-(4-formylphenoxy)-N-arylacetamides with 1,3-indandione produced good yields of the desired products. The structural features of the novel compounds were validated using spectrum and elemental analysis. The cytotoxic effect of the prepared compounds were determined against several human cancer and normal cell lines. It was found that compounds 5d and 5e exerted the most promising cytotoxic activity against Hela cells with IC50 (4.8 and 5 μg/mL, respectively) relative to doxorubicin (5 μg/mL). The molecular studies showed that compounds 5d and 5e decreased the expression level of Antigen kiel 67 (KI-67), Protein kinase B (AKT), Survivin, Interleukin-6 (IL-6), Interleukin-1beta (IL-1B), and Cyclooxygenase-2 (COX-2) genes in Hela treated cells. In addition, the activity of Glutathione (GSH), Caspase-9, Caspase-8, Caspase-3, P53, and BAX were significantly enhanced and Bcl2, Malondialdehyde (MDA), and IL-6 were suppressed in 5d and 5e treated Hela cells. Compounds 5d and 5e arrested the cell cycle at G0/G1 phase and decreased the migration ability of Hela cells to close the scratch gap after 48 h. Molecular docking study supported our ELISA results, where compounds 5d and 5e showed promising binding energy scores against P53 cancer mutant Y220C and Bcl2.

Summary, etc. لقد تم إنشاء سته شالكونات جديدة، وتم استخدام العديد من التقنيات الطيفية لتحديد بنيتها. لقد تم اختبار كل مركب مُصنَّع للتأكد من سميته للخلايا ضد العديد من خطوط الخلايا السرطانية، وأظهر مركبات 5d & 5eافضل النتائج ضد HELA. لقد اوضح تفاعل البوليميراز المتسلسل للنسخ العكسي أن استخدام مركب 5d & 5e له تأثير مثبط على مستويات التعبيرالجينى لجينات ki67، وsurvivin، وIL-1B، وIL-6، وCOX-2، وAKT1. لقد قاطع المركب 5d & 5e دورة الخلية في مرحلة G0/G1 في الخلايا المعالجة HELA على التوالي وفقًا لتحليل التدفق الخلوي لدورة الخلية. تم ملاحظه زياده نشاط Caspase 8،Caspase 9 ، P53، BAX و GSH بشكل كبير وتثبيط ، BCL2، MDA و IL-6 في خلايا HELA المعالجة بـ 5d & 5e وفقًا لفحص ElISA (المقايسة الامتصاصية المناعية للانزيم المرتبط ). و أظهرفحص التئام الجروح بعد 48 ساعة من المعالجه أن شالكون 5d & 5e قلل من عدد خلايا HELA المهاجرة الي مكان الجرح والقدرة على إغلاق جرح الكشط مقارنةً بالخلايا الغير المعالجة. لقد تم التحقق من صحة بيانات الاليزا الخاصة بنا من خلال النمذجة الجزيئية النظرية ضد طفرات سرطان P53 Y220C وBCL2.
