Summary, etc. Introduction Tecoma stans (T. stans) is a fast-growing plant from the <br/>family Bignoniaceae. Tecoma stans (L.) Jus., a prevalent tree in Egypt, was <br/>reported to be a promising photosensitizer in PDT.Various parts of T. stans <br/>have been used in different biological applications, especially in cancer <br/>treatment and antimicrobial activities. Photodynamic therapy (PDT) is a <br/>promising modality for cancer treatment and antimicrobial activities that <br/>depends on the interaction between a photosensitizer, light, and oxygen. <br/>Tecoma stans yellow flowers crude cold methanolic extract (TSFE) is a <br/>promising anticancer, antimicrobial, and natural photosensitizer for PDT and <br/>this study may inspire further ethnobotanical investigations. <br/>Aim Searching for photosensitizers from plant origin is crucial to <br/>providing nontoxic photosensitizers with high economic antineoplastic and <br/>antimicrobial value. <br/> Method In this study, we investigated the photodynamic activities of <br/>(TSFE) as a natural photosensitizer (PS) source. The phytoconstituents of <br/>TSFE were analyzed by the UPLC/MS/MS technique. This technique <br/>revealed that the extract held compounds that possess cytotoxic and <br/>antimicrobial activities, like luteolin, kaempferol, and isorhamnetin. <br/>Moreover, the photosensitizing agents like coumarins and anthocyanins. The <br/>cytotoxicity of TSFE was examined on the breast carcinoma (MCF-7) and <br/>lung carcinoma (A549) cell lines, in the dark and after irradiation by blue <br/>light (400-450 nm). TSFE contained various phytochemical components with <br/>antineoplastic activity. Moreover, (TSFE) contains coumarins and <br/>anthocyanins that may act as photosensitizers. Furthermore, we investigated <br/>the antimicrobial photodynamic activities of TSFE as a natural <br/>photosensitizer (PS) source against microbes. TSFE was first prepared in a <br/>nanoemulsion form to enhance bioavailability. Antimicrobial photodynamic <br/>therapy (aPDT) is an approach that was recently used to combat the <br/>resistance developed by pathogens toward antimicrobial drugs. This study <br/>investigates the antimicrobial photodynamic activity of the methanolic crude <br/>cold extract of Tecoma stans (L.) Juss. ex Kunth loaded in a nano-emulsion <br/>as a natural photosensitizer. Molecular docking and molecular dynamics <br/>studies were conducted to investigate the binding between delphinidin as a <br/>promising lead and DNA's active site and assessed its stability through <br/>simulation. The nano-emulsion formulation (TSFE-NE) was prepared, <br/><br/><br/>Abstract <br/><br/> <br/><br/>X <br/><br/> <br/><br/>characterized, and evaluated for its antimicrobial photodynamic activities <br/>against three types of microorganisms: Gram-negative bacteria Escherichia <br/>coli (E. coli), Gram-positive bacteria Staphylococcus aureus (S. aureus), and <br/>the pathogenic yeast Candida albicans (C. albicans). <br/> Result )TSFE( showed negligible cytotoxicity against MCF-7 cell lines <br/>at all tested concentrations in the dark. A non-significant cell viability change <br/>was observed upon radiation (P>0.05). TSFE showed significant dark <br/>cytotoxicity on A549 cells, which improved significantly after light radiation <br/>(P<0.05).The percentage of microbial eradication was calculated after <br/>irradiation at 90 Mw/cm2. The microbial eradication of TSFE-NE was <br/>enhanced after light exposure. Eradication percentages of 97.43%, 90.56%, <br/>and 87.77% were obtained for Escherichia coli, Staphylococcus aureus, and <br/>Candida albicans, respectively. These results confirmed the efficacy of <br/>natural photosensitizer and nanotechnology combination for improving <br/>antimicrobial photodynamic activities .
