| Summary, etc. | Coragen 20 SC is a new insecticide contains the active ingredient <br/>chlorantraniliprole, has been approved for use against insects in many crops due <br/>to the presence of insect ryanodine receptors. Coragen is being used to fight <br/>various types of flies and their larvae. The present study aims to evaluate the <br/>intrauterine exposure to coragen in pregnant rats and their fetuses from 6th to <br/>20th day of gestation. Forty-five albino rats (Rattus norvegicus); 15 males and 30 <br/>virgin females, weighing 170 ± 20 g were provided from the animal house of the <br/>National Research Center, Egypt. The pregnant dams were orally administrated <br/>saline (control), 100 mg/kg (low dose) or 200 mg/kg (high dose) of coragen. <br/>Pregnant rats were checked daily to determine any signs of toxicity or <br/>behavioral change. The weights of the pregnant rats were weekly recorded. The <br/>mothers of two treated groups showed teratogenic anomalies such as <br/>asymmetrical distribution of fetuses in the two uterine horns, as well as, <br/>observed resorption, bleeding in the uteri while, their fetuses had growth <br/>retardation, morphological malformations and some skeletal perversions in <br/>addition to some dead fetuses were recorded. Fetal and maternal specimens of <br/>liver and kidney of the control and treated groups were firstly fixed in 10% <br/>buffered formalin solution for 24 hrs., then, dehydrated using ascending series <br/>of ethyl alcohol. Specimens were cleared in xylol and embedded in paraffin <br/>wax. The samples were sectioned at 5 μm and stained with H&E for routine <br/><br/><br/> <br/><br/> <br/><br/>examination and photomicrography by the light microscope. Furthermore, <br/>multiple histopathological changes occurred, and a significant DNA damage <br/>was induced by coragen in maternal and fetal liver and kidney tissues compared <br/>to the control. Gestational coragen treatment resulted in teratogenic and <br/>genotoxic effect on pregnant rats and their fetuses. The obtained results in the <br/>present study imply that women and their fetuses may have the same risk. |

| Summary, etc. | الكوراجين 20 إس سي هو مبيد حشري جديد يحتوي على المادة الفعالة كلورانترانيليبرول، وقد تمت الموافقة على استخدامه ضد الحشرات في العديد من المحاصيل بسبب وجود مستقبلات الريانودين الحشرية. يستخدم الكوراجين لمحاربة أنواع مختلفة من الذباب ويرقاته. تهدف الدراسة الحالية إلى تقييم التعرض داخل الرحم للكوراجين في الجرذان الحوامل وأجنتها من اليوم السادس إلى اليوم العشرين من الحمل. خمسة وأربعون من الفئران البيضاء (Rattus norvegicus)؛ تم توفير 15 ذكراً و30 أنثى ، بوزن 170 ± 20 جم من بيت الحيوان التابع للمركز القومي للبحوث بمصر. كانت الفئران الحوامل تجرع عن طريق الفم بمحلول ملحي (تحكم)، 100 ملغم/كغم (جرعة منخفضة) أو 200 ملغم/كغم (جرعة عالية) من الكوراجين. تم فحص الفئران الحوامل يوميًا لتحديد أي علامات سمية أو تغير سلوكي. تم تسجيل أوزان الجرذان الحوامل اسبوعيا. أظهرت أمهات المجموعتين المعالجتين تشوهات مثل التوزيع غير المتماثل للأجنة في قرني الرحم، وكذلك ملاحظة اختفاء بعض الاجنه ونزيف في الرحم بينما كانت الأجنة تعاني من تأخر النمو والتشوهات للشكل الخارجى وبعض التغيرات الهيكلية بالإضافة إلى تسجيل بعض الأجنة الميتة. تم أولاً تثبيت عينات الكبد والكلى الجنينية والأمية للمجموعات التحكم والمعالجة في محلول الفورمالين المنظم بنسبة 10% لمدة 24 ساعة. ثم يتم تجفيفه باستخدام سلسلة تصاعدية من الكحول الإيثيلي. تم مسح العينات في الزيلول ودمجها في شمع البارافين. تم تقسيم العينات عند 5 ميكرومتر وتصبغ بـصبغة H&E للفحص الروتيني والتصوير المجهري الضوئي بواسطة المجهر الضوئي. علاوة على ذلك، حدثت تغيرات نسجية مرضية متعددة وحدث تلف كبير في الحمض النووي بواسطة الكوراجين في أنسجة الكبد والكلى للام والاجنه مقارنة بالتحكم. أدى علاج الكوراجين للفئران الحوامل إلى تشوهات الاجنه وتغيرات جينيه على الجرذان الحوامل وأجنتها. تشير النتائج التي تم الحصول عليها في هذه الدراسة إلى أن الامهات وأجنتهن قد يتعرضن لنفس المخاطر. |
