Dissertation note: Thesis (Ph.D.) - Cairo University - Faculty of Science - Department of Botany and Microbiology Summary: In this work, the microfloral picture from Saint Catherine protectorate (SCP), Giza zoo garden (GZG) and Cairo university (CU) soils was studied. Twenty five fungal isolates, six bacterial isolates were isolated. Aspergillus flavus isolated from saint Catherine protectorate (SCP) was the most active organism in laccase production. The culture medium was optimized for laccase production. Laccase was further purified and characterized, it showed an apparent molecular weight of 68.5 kDa by SDS-PAGE. Aspergillus flavus purified laccase was capable of decolorizing seven dyes of concentration with different degrees, the highest and most rapid decolorization occurs with malachite green. Laccase tested on malachite green decolorization with coexistence of metal ions and organic solvents and was also tested on real textile effluent over 6 days. Aspergillus flavus was deposited in NCBI Gen Bank and was given a strain identifier, A. flavus NG85. Aspergillus flavus NG85 putative laccase gene was isolated, sequenced, with accession number KM522917.2
