As a prelude to developing a yeast - based fermentation process for the production of phenylalanine free alpha - casein as a foodstuff for patients suffering from phenylketonuria , we cloned a gene encoding a nutritional protein (bovine alpha - casein) , but devoid of phenylalanine codons and with a codon bias similar to that of naturally occurring highly expressed genes in ScerevisiaeThe results show that synthetic gene is expressed in Ecoli when cloned into the Ecoli expression vector pET28a , and expressed in Scerevisiae when cloned into yeast expression vector pYES2 , Western blot analysis has confirmed the expression of the gene understudy conclusion : We cloned and expressed a gene devoid of any phenylalanine codon to produce a foodstuff for PKU patients
