Samples (water & O. niloticus) were collected from the river Nile at El- Qanater El- Khyria as non-polluted site and from El-Rahawy drain region as a polluted area during 2016, seasonally, for measuring the physico-chemical characteristics of water, measuring of some heavy metals in water and fish (Fe, Zn, Cu, Pb, Mn and Cd). The obtained results revealed changes in physico-chemical parameters that have negative impacts on water quality and other changes in heavy metals concentration in water and fish of the studied areas. Electrophoretic pattern of the native protein, calcium moieties and Ý-esterase for fish liver and muscles was used as Bio-indicator of water pollution. The values of hazard index (HI) and hazard quotient (HQ) of El-Qanater samples are lower than the acceptable limits, while HI and HQ for the analyzed heavy metals were higher than the acceptable limit at El-Qatta station. Accordingly, the metals cumulative risk effects gave an alarming sign specifically at high fish consumption rates at El-Qatta station
