Dissertation note: Thesis (M.Sc)-Cairo University, 2025. Summary: The present study was carried out in the apiary yard of the Faculty of Agricultural, Cairo University, Giza, aiming to detect the suitable conditions with respect to colony’s queen state and drones age (1-3 & 7-9 & 12-14 & 21-23 days old) for obtaining maximum values of some reproductive characteristics of honeybee drones. In addition, to detect the suitable method for introducing honeybee queens after instrumental insemination for obtaining high number of sperms in the spermatheca combined with minimum residues of semen in the lateral oviducts. The results showed that the number of spermatozoa in drones significantly decreased with advancing age. It was also found that the number of spermatozoa in drones raised in the presence of a queen was significantly higher than those raised in the absence of a queen, regardless of the ages of the drones raised. The results also clarified that the volume of semen in drones raised in the presence of the queen was greater than the volume of semen in drones raised in the absence of the queen. The results also showed that the percentage of mature drones producing semen was significantly higher when reared in the presence of the queen compared to those reared in the absence of the queen. In addition, the instrumentally inseminated queens being placed under semi-spherical screen cages with 15 workers showed significant decrease in the number of sperms in the spermatheca in comparison with the other treatments. Also, the results showed that the number of sperms in the spermathecae of the queens placed under semi-spherical queen excluder cages or introduced in mini nuclei was significantly higher than that of the queens introduced under semi-spherical screen cages with 15 workers. The results also showed that the highest number of spermatozoa was found in the spermatheca of queens that were instrumentally inseminated and placed in mating nuclei. In addition, the results indicated that the number of spermatozoa in the spermatheca of queens that were naturally mated was slightly fewer than the number of spermatozoa in the spermatheca of queens that were instrumentally inseminated and placed in mating nuclei, or under semispherical queen excluder cages. but these differences were not significant. In contrast, the number of sperm retained in the lateral oviducts of the instrumentally inseminated queens was significantly higher in queens introduced under semi-spherical screen cages with 15 workers than those introduced under semispherical queen excluder cages, or in mating nuclei.
