520			_aArthritis is one of the most important diseases causing lameness in dogs as well as other species of animals. Osteoarthritis (OA) cause considerable deterioration of the cartilage and marked enzymatic changes in the synovial fluid. The present study was conducted on 25 clinically healthy male Mongrel dogs, with body weight of 15.0 ±1.7kg. Arthritis was induced in 20 dogs and 5 dogs were left as a negative control (normal) group. Osteoarthritis was induced by performing arthrotomy and curettage of the cartilage by creation of two 8.0 mm diameter full thickness chondral defect till exposing the subchondral bone. The dogs were left for 10 days till complete healing of the surgical wound. Amphotericin-B was injected intra- articularly at a dose of 10 mg weekly for 2 successive weeks. The dogs were left one week till complete typical symptoms of arthritis were observed. The experimental groups were classified into five groups as control negative (normal), dexamethasone treated, sodium hyaluronate treated, poly sulfated glycosaminoglycan treated and control positive (sham operated). Sensory characteristics of the treated groups were evaluated. Besides, synovial analysis was Matrixweeks.th 4,rd 3,nd 2,st 1theofendtheatlperformed for each anima metalloproteinase (MMPs) 3 and 13 and tissue inhibitor metalloproteinase (TIMP- 1) were detected by ELISA assay. The obtained results revealed that osteoarthritis OA was associated with significant increases in the enzymatic activities of MMPs 3 and 13 and decrease of TIMP-1 as biomarkers of OA. Among the used treatments, polysulfated glucose aminoglycan is the highly recommended therapeutic approach as it caused the highest improvement compared with other treatments.
520			_aالتهاب المفاصل هو واحد من أهم الأمراض التي تسبب العرج في الكلاب وكذلك الأنواع الأخرى من الحيوانات . يسبب التهاب المفاصل (OA)تدهورا كبيرافي الغضروف وتغيرات انزيمية ملحوظة في السائل الزليلي والأنسجة المشتركة . أجريت هذه الدراسة على 25 كلبا ذكرا يتمتعون بصحة جيدة سريريا ، ويبلغ وزن الجسم 15.0 ±1.7 كجم .تم تحفيز التهاب المفاصل في 20 كلبا وترك 5 كلاب كمجموعة تحكم سلبية ( طبيعية ) . تم تحفيز التهاب المفاصل عن طريق اجراء فتح المفاصل وكشط الغضروف عن طريق انشاء اثنين من عيوب الغضروف بقطر 8.0 ملليمتر كامل السماكة حتى تعريض العظم تحت الغضروف . تركت الكلاب لمدة 10 أيام حتى الشفاء التام للجرح . تم حقن الأنفوترسين-بي داخل المفصل بجرعة 110 ملليغرام مره أسبوعيا لمدة أسبوعين متتاليين . تركت الكلاب أسبوعا واحدا حتى لوحظت الأعراض النموذجية الكامله لإلتهاب المفاصل . تم تصنيف المجموعات التجريبية إلى خمس مجموعات على انها - سلبية التحكم (طبيعية )2- ديكساميسازون المعالجة 3- هيالورينات الصوديوم المعالجة 4- جلوكوز أمينو جليكان متعدد الكبريتات المعالجة 5- إيجابية ( بدون علاج تعمل كمجموعة ضابطة ) . تم تقييم الخصائص الحسية للمجموعات المعالجة إلى جانب ذلك، تم إجراء التحليل الزليلي لكلٍ في نهاية الأسابيع الأول والثاني والثالث والرابع . تم الكشف عن مصفوفة MMPs 3,13 ومثبطات الأنسجة TIMP-1 بواسطة فحص إليزا . كشفت النتائج التي تم الحصول عليها ان التهاب المفاصل OAارتبطت بزيادات كبيرة في الأنشطة الإنزيمية ل MMPs3,13وانخفاض TIMP-1 كمؤشرات حيوية ل OA. من بين العلاجات المستخدمة ، يعد الجلوكوز أمينو جليكان متعدد الكبريتات هو النهج العلاجي الموصى به للغاية لأنه تسبب في أعلى تحسن مقارنة بالعلاجات الأخرى .
