Dendritic cells (DCs) are professional antigen-presenting cells, owing to their superior capacity for acquiring and processing antigens for presentation to T cells. Since dendritic cells were identified as master cells in the modulation of the immune response, a lot of effort has been made to bring their potential to the clinical setting. Dendritic cells - based vaccines are considered an attractive tool for cancer immunotherapy that aim to reset a patients immune system and thereby elicit anti-tumor responses in vivo. In general, generation of MDDC (monocyte derived DC) from monocytes is induced in the presence of GM - CSF and IL - 4, and a maturation stimulus is added to the culture. TNF - Ü is the most common cytokine used for activating DC and generating mature MDDC. A chemically modified Lipopoly-saccharide derivative, monophosphoryl lipid A (MPLA), is one of the promising toll like receptor (TLR) ligands. It has been safely used as an adjuvant in various vaccine trials in humans. In our study, we compared the effects of traditional cytokine coktail (TNF alpha + IL - 1Ý) and MPLA as different maturation stimuli on MDDC
